摘 要:目的:建立首乌藤配方颗粒的HPLC指纹图谱,并对其主要的共有峰进行成分归属,为其质量控制提供科学依据。方法:采用HPLC方法,测定了不同厂家的15批首乌藤配方颗粒的HPLC指纹图谱,色谱条件:Waters XBridge^(TM)shield RP18 C_(18)色谱柱,流动相乙腈-0.1%乙酸水溶液,流速1.0 m L·min^(-1),柱温30℃,检测波长271 nm,进样量15μL。结果:建立了首乌藤配方颗粒的指纹图谱,采用"中药色谱指纹图谱相似度评价系统"计算处理,通过HPLC指纹图谱分析,确定了首乌藤配方颗粒8个共有峰,并确认6个特征峰,分别为没食子酸,原儿茶酸,儿茶素,2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷,大黄素,大黄素甲醚。对照指纹图谱色谱峰丰富,分离度高,匹配确定了主要特征峰,15批首乌藤配方颗粒样品指纹图谱与对照指纹图谱相似度均>0.86,相似度高,可实现首乌藤配方颗粒化学成分全面和整体的评价,能较好控制首乌藤配方颗粒的质量。结论:该方法简单、准确、重复性好,可以为首乌藤配方颗粒质量控制和评价提供参考。Objective: To establish a method of fingerprint analysis on Polygoni Multipiori Caulis formula granules by HPLC,identify the main chemical compounds,and provide a reference for their quality control.Method: The HPLC method was used to determine the HPLC fingerprint of fifteen batches of Polygoni Multipiori Caulis formula granules from different manufacturers. The analysis was performed on a Waters XBridge^(TM)shield RP18 C(18) column with acetonitrile-0. 1% acetic acid solution as the mobile phase for gradient elution. The flow rate was 1. 0 m L·min-1; the detection wavelength was 271 nm; the column temperature was 30 ℃; and the injection volume was 15 μL. Result: The fingerprint of Polygoni Multipiori Caulis formula granules was established,and the results were calculated according to Similarity Evaluation System for Traditional Chinese Medicine Chromatographic Fingerprint. With the analysis of the HPLC fingerprints,totally eight common peaks were found,including six identified peaks for gallic acid, protocatechuic acid, catechin, 2, 3, 5, 4'-tetrahydroxystilbene-2-O-β-Dglucoside,emodin,and physcion. Good similarities were found in the established fingerprint through similarity analysis and the resolution was good. Main characteristic peaks were also determined,and the similarities of the chromatograms were all larger than 0. 86,which can realize comprehensive and overall evaluation of its chemical compositions and better control the quality. Conclusion: The method is simple and accurate with good reproducibility,which may provide the reference for quality control and evaluation of Polygoni Multipiori Caulis formula granules.
