摘 要:通过组织培养建立乌菜离体再生体系,可使优良珍贵育种材料得到保存,并能为其快速繁殖提供有效途径。本研究以4种不同基因型乌菜为材料,从外植体类型、基本培养基类型、植物生长调节剂配比、基因型及无菌苗苗龄等方面,对影响乌菜不定芽诱导的因素进行了研究。研究表明,7-1基因型乌菜5 d苗龄的带柄子叶外植体,在以MS为基本培养基(3%蔗糖+0.8%琼脂)添加4 mg/L 6-BA与0.5 mg/Lα-NAA的诱导培养基上进行诱导,不定芽诱导率可达85.56%。本研究对影响乌菜植株再生的多个因素进行了优化,建立了高效离体再生技术体系,为乌菜育种研究及其他生物技术研究提供基础。By the in vitro regeneration system of Wucai, the valuable breeding materials could be saved and propagated rapidly. To establish this regeneration system, 7-1, 3-5, Wl8 and T98-9, four different genotypes of Wucai were used as experimental materials in this study. Several factors affecting the adventitious shoots induction, including explant type, basic media, plant growth regulators concentration, genotype and seedling age, were analyzed and optimized. When the cotyledons with petiole from 5-day seedlings of Wucai genotype 7-1 were inoculated as the explant on the induced medium (MS medium supplemented with 3% sucrose, 0.8% agar, 4 mg/L 6-BA and 0.5 mg/L α-NAA), the regeneration rate of adventitious shoots reached up to 85.56%. In this study, the factors affecting in vitro regeneration of Wucai were optimized, and an effective regeneration system of Wucai cotyledons with petiole had been established, which could provide the foundation for Wucai breeding and other biotechnology researches.
