摘 要:如今泥炭藓植物的使用受到越来越多的重视,导致人工采摘严重,泥炭藓湿地严重破坏,本研究针对该问题进行泥炭藓植物的人工培养试验,以多纹泥炭藓外植体为实验材料,筛选出无菌苗存活率最高,染菌率最低的消毒方法:200 mg/L的青霉素浸泡2.5 h,75%酒精浸泡60 s,0.3%浓度下的次氯酸钠浸泡60 s,在该消毒方法下以Konp培养基为基本培养基,多纹泥炭藓存活率达100%,染菌率为33%;将无菌苗接入Beneck、MS、BG-11和Knop 4种培养基内进行组织培养,筛选出最适培养基-Beneck培养基;在不同培养基中加入10个不同浓度梯度的蔗糖,筛选出发芽率最多的培养基及其最适蔗糖浓度,即Beneck培养基,蔗糖浓度为2%。最适的消毒配方和培养基配方将会在短期时间内提供大量的多纹泥炭藓无菌苗,对解决添加栽植苗供应量不足的问题起到积极帮助。Nowadays, the usage of Sphagnum plants are getting more and more attention, which leading to the serious manual pluck, and sphagnum wetlands were severely damaged. Refer to this condition, we studied the artificial cultivation of Sphagnum plants, picked the explant of Sphagnum multifibrosum as experimental material, selecting the disinfection method with survival rate of aseptic seedling and the lowest infection rate. We soaked the explants into 200 mg/L penicillin with 2.5 h, 75% alcohol with 60 s and 0.3% concentration of sodium hypochlorite with 60 s. Based on this disinfection method, we used Konp as basic culture medium, the survival rate of Sphagnum multifibrosum reached 100% and the contamination rate was 33%; Then we vaccinated the aseptic seedling into 4 mediums (Beneck, MS, BG-11 and Knop) for tissue culture, selected the optimum culture medium-Beneck culture medium. Fina.lly, we added 10 kinds of different concentration gradient in different culture medium, selecting medium with the highest germination rate and its optimum sucrose concentration, which was Beneck and sucrose concentration of 2%. The optimum disinfection formula and medium formula would provide a large number of the Sphagnum multifibrosum aseptic seedling in a short period oftime and give a positive help for filling the lack of seedling demand in field planting.
