肺结核患者外周血不同T淋巴细胞亚群中白细胞介素-9产生细胞比例的研究

摘　　要：目的探讨肺结核病（tuberculosis，TB）患者外周血不同T淋巴细胞亚群中白细胞介素（interluekin，IL）-9产生细胞比例的变化。方法采集26例活动期肺TB患者（TB组）和23例健康志愿者（HD组）外周血样本，加刺激剂佛波醇酯（PMA）和钙离子霉素（IM），以及阻断剂莫能霉素作用6h后，用荧光标记单克隆抗体作T细胞亚型表面分子和胞内细胞因子染色，流式细胞仪检测不同T淋巴细胞亚群中分泌IL-9细胞的比例。结果CD3＋总T细胞中产生IL-9的细胞比例在TB组和HD组分别为0．32％和0．35％，在CD4＋αβT细胞中分别为0．21％和0．22％，在CD8＋αβT细胞中分别为0．35％和0．32％，差异均无统计学意义（P〉0．05）。而在γδT细胞中产生IL-9细胞的比例，TB组（2．93％）高于HD组（1．07％），差异有统计学意义（P〈0．05）。结论与HD组比较，TB患者CD4＋T细胞和CD8＋T细胞中IL-9产生细胞的比例无明显改变，而18T细胞中分泌IL-9细胞比例显著增加。Objective To explore the changes of the proportion of （interleukin, IL）-9 producing T cells among different T cell subpopulations of peripheral blood from patients with active pulmonary tuberculosis （TB）. Methods Peripheral blood samples were obtained from 26 pulmonary TB patients and 23 health donors,and stimulated with Phorbo 12-myristate 13-acetate （PMA） and ionomycin （IM） for 6 h in the presence of monensin （Mo） at last 4h. Cells were collected and stained with fluorochrome conjugated mAbs, to cell surface makers for phenotypes and to intracellular cytokine. The proportions of IL-9 producing cells in different T cell subpopulations were detected by flow cytometry. Results The proportions of IL-9 producing cells among total CD3 ＋T cells in TB patients and health donors were 0.32%, and 0. 35 %, respectively, and that among CD4 ＋ αβ T cells were 0.21% and 0.22%, respectively;and that among CD8 ＋ αβ T cells were 0. 35% and 0. 32% respectively ; there were no significant differences in two groups. Whereas, the percentage of IL-9 producing cells in γδ T cells in TB patients （2.93%） was significantly higher than that in healthy donors group（ 1.07% ） （P 〈 0.01 ）. Conclusion Compared to healthy donors group, there is no significant change of IL-9 producing cell proportions in CD4 and CD8αβ T ceils in TB patients. However,there is increase of IL-9 producing γδT cells in peripheral blood of TB patients.
