摘 要:为了研究山西地区猪流行性腹泻病毒M基因的遗传变异情况,从2014年10月~2015年4月山西地区11个地市收集的猪流行性腹泻病毒(PEDV)病料中扩增到4株PEDV的M基因,并进行序列比对及遗传分析。结果表明,4株PEDV的M基因与CV777毒株比较,部分核苷酸及氨基酸发生改变。4株PEDV的M基因核苷酸和氨基酸的同源性分别为99.6%~100%和99.1%~99.6%,与参考毒株的核苷酸和氨基酸的同源性分别为96.3%~99.9%和96.0%~99.1%。遗传进化分析显示,4株PEDV与2010-2013年中国分离株(BJ2010、HB/BD、HB/FN、HLJ-2012、GDHZ-1202、SHQP/YM/2013)、2008年泰国KU04RB08株亲缘关系较近;与2株泰国株(M_NIAH2013_95和M_NIAH1795_04)、2株欧洲株(CV777和Br1/87)和2006年中国分离株LZC亲缘关系较远。In order to study the heritable variation of M gene of porcine epidemic diarrhea virus( PEDV) in Shanxi Region,4PEDV field strains were isolated from 11 cities in Shanxi province during October 2014 to April 2015. M gene was amplified from the4 PEDV field strains by RT-PCR,cloned,sequenced and compared with the other reference strains in Gen Bank. The result showed that the several nucleotides and amino acids of M gene of 4 PEDV field strains compared with the vaccine strain( CV777) changed.The nucleotide and amino acid homologies of M gene of the 4 PEDV field strains in Shanxi were 99. 6% to 100% and 99. 1% to99. 6%,respectively,but compared with the reference strains were 96. 3%-99. 9% and 96. 0%-99. 1%. Phylogenetic analysis showed that the 4 PEDV strains had closer genetic relationship with the Chinese isolates( BJ2010,HB/BD,HB/FN,HLJ-2012,GDHZ-1202,SHQP/YM/2013) during 2010 to 2013 and Thailand isolate( KU04RB08) in 2008,but more distant genetic relationship with European isolate( CV777,Br1/87),Thailand isolates( M_ NIAH2013_95,M_ NIAH1795_04) and China isolate( LZC) in 2006.
