摘  要:为分析肠炎沙门菌感染后不同时间点济宁百日鸡盲肠TLR2、TLR4、NOD1和NALP3基因表达的变化规律,试验选用2日龄肠炎沙门菌阴性济宁百日鸡为试验动物,接种肠炎沙门菌,应用荧光定量PCR分别检测接种后第1、3、7、14、21、28和35天试验组与对照组盲肠组织TLR2、TLR4、NOD1、NALP3 4个基因的表达变化。结果:肠炎沙门菌感染后第3、7、21、28和35天,试验组和对照组盲肠TLR2的表达量差异显著(P<0.05),除感染后第3天TLR2的表达量显著降低外,其他各时间点TLR2的表达量均显著升高(P<0.05);感染后第3、21、28天TLR4的表达量显著升高,分别为对照组的1.79、1.52、1.21倍(P<0.05);感染后第7天,试验组NOD1和NALP3的表达量均显著低于对照组,分别为对照组的0.37和0.61倍(P<0.05);感染后第21天,试验组NALP3基因的表达量显著升高,为对照组的1.27倍(P<0.05)。研究结果表明:TLRs和NLRs基因在肠炎沙门菌感染后被激活,且表现出协同调控作用,这种协同调控在感染后第7天最为明显。In order to study the expression of TLR2, TLR4,NOD1 and NALP3 genes in Jining Bairi chicken cecum following Salmonella enteritidis (SE)infection, the two-day-age SE free Jining Bairi chickens were selected and infected with SE. On the day 1,3,7,21,28 and 35 post infection, cecum samples were collected for RNA extraction. Quantitative PCR was used to analyze the expression of TLR2, TLR4, NOD1 and NALP3 by SYBR Green I method. The results showed that on the day 3,7,21,28 and 35 after SE infection, TLR2 was significantly differentially expressed (P〈0.05).The TLR2 was significantly down-regulated on day 3 after infection,but up-regulated on the other time points after infection (P〈0.05). The expression level of TLR4 in treated group was 1.79, 1.52,and 1.21 fold change higher than that in control group on day 3,21 and 28 after SE infection,respectively (P〈0.05). NOD1 and NALP3 were significantly down-regulated lower than those in the control group with the fold change of 0.37 and 0.61, respectively, on day 7 after infection (P〈0.05). The NALP3 was significantly up-regulated in the treated group than that in the control group with the fold change of 1.27 on day 21 after infection (P〈0.05). This study revealed that the TLRs and NLRs genes were activated after SE infection and showed co-regulation. The co-regulation was the most obvious on day 7 after SE infection.
