日本血吸虫可溶性虫卵和童虫抗原刺激RAW264.7巨噬细胞的初步分析

摘  要:本文采用不同浓度的日本血吸虫虫卵抗原(SEA)、童虫抗原(SSA)刺激小鼠单核巨噬细胞RAW264.7,结果显示SEA抗原能明显刺激巨噬细胞增殖分化(SI〉2),且多数巨噬细胞形态发生了改变并出现细胞死亡。实时定量荧光PCR结果显示,在SEA、SSA抗原刺激下,巨噬细胞TNF-α、IL-6、IL,10和CCL2的mRNA表达量都明显高于对照组。结果提示血吸虫抗原对巨噬细胞的刺激作用可能在宿主抗血吸虫感染的先天免疫应答和虫卵肉芽肿形成免疫调节中发挥了一定的作用。In this study, we stimulated mouse macrophages RAW264.7 using different concentration of soluble schistosome egg antigen (SEA) and schistosomulum antigen (SSA) respectively, the observations showed that SEA could significantly stimulate the proliferation and differentiation of RAW264.7 cells (SI 〉 2) , and the morphology of the cells were changed dramatically and some of them were died. qPCR analysis revealed that the expression level of TNF-α, IL-6, IL-10 and CCL2 were significantly higher in SEA or SSA stimulated cell groups than that of the control group. The stimulation effects of macrophages by schistosome antigens may play an important role in the regulation of host's innate immune response and granuloma formation in schistosomiasis.
