摘 要:目的探索抑癌基因PTEN在子宫内膜异位症(EMs)发病中的作用,观察痛经宁对EMs大鼠在位与异位内膜组织中PTEN蛋白及mRNA表达的影响。方法自体移植法建立EMs大鼠模型,随机数字表法分5组,分别给予孕三烯酮与高、中、低剂量的痛经宁药液及蒸馏水对照。28天后,通过HE染色观察各组大鼠在位与异位内膜组织的病理结构;免疫组化(S-P)法、免疫印迹(Western Blot)法测定PTEN蛋白的表达情况;qPCR法测定PTENmRNA的表达情况。结果在位及异位内膜组织中,EMs大鼠PTEN蛋白及mRNA呈低表达(P<0.05),异位内膜低于在位内膜。高剂量的痛经宁药液使之表达强度上升最多,差异具有统计学意义(P<0.05)。病理结构显示,各剂量的痛经宁药液均可使异位内膜萎缩,以高剂量组最为明显,不改变在位内膜的结构。而孕三烯酮可使在位及异位内膜均萎缩。结论 PTEN的降低是EMs的发病机制之一,痛经宁可通过上调PTEN蛋白及mRNA的表达而起效,可使异位内膜出现凋亡性病理改变,而不影响在位内膜的病理结构。Objective To explore the role of PTEN in the pathogenesis of endometriosis ( EMs ) and to observe the efl'ect of Tongjingning on the expression of PTEN protein and mRNA in eutopic and ectopic endometrium tissue of rats with EMs. Methods We built EMs model of rats using their own endometrial tissue, then divided the successful model of rats into Tongjingning low, middle, high dose group, Gestrinone group and distilled water group in accordance with the random number table. After 2g days' treatment, we observed the pathological structure of eutopic and ectopic endometrium tissue in all groups by HE staining; we de- tected the expression of PTEN protein by immuno histochemical (S- P) and immuno blot (Western Blot) ; we detected the ex- pression of PTENmRNA by qPCR. Results The PTEN protein and mRNA were lowly expressed in eutopic and eetopic endometri- um tissue of rats with EMs (P 〈 0.05). The expression was lower in ectopic endometrium than eutopic. The expression in Tongjingning high dose increased most and the difference was statistically significant ( P 〈 0.05 ). The pathological structure showed that each dose of Tongjingning can make the ectopic endometrium tissue atrophied. The high dose was the most obvious which didn' t change the structure of eutopic endometrium tissue. Gestrinone could make both eutopic and ectopic endometrium atrophied. Conclusion The decrease of PTEN is one of the pathogenesis of EMs and Tongjingning can cure it by up - regulating the expression of PTEN protein and mRNA. The apoptosis only exsits in the ectopic endometrium not the eutopic.
