摘　　要：为了探索克雷伯氏杆菌的重金属抗性机理,提取Ag+、Pb2+和Cr3+胁迫后克雷伯氏杆菌的胞内蛋白,利用双向电泳技术和质谱技术分离、解析、鉴定特异性蛋白质斑点。结果表明,不同重金属离子胁迫后克雷伯氏杆菌的蛋白质斑点在数量和表达量上有较大的差异,但存在3个表达新增和10个表达关闭的共同蛋白斑点,质谱鉴定结果表明特异表达蛋白质与抗氧化系统、能量代谢调节机制有关。研究表明,克雷伯氏杆菌受到重金属胁迫后启动过氧化氢酶的表达,同时通过关闭胞内原有碳分解代谢途径、开启磷酸戊糖途径等相应的应急系统获取能量,降低重金属离子对细胞的毒害作用。This paper is entitled to provide a mechanistic insight into the toxicity and a counter defense system of antioxidants in Klebsiella pneumonia.As a matter of fact,heavy metals,such as lead（Pb）,chrome（Cr） and silver（Ag）,are common contaminants and tend to be inherent in the environment and bring about biological threats to human beings.But,on the contrary,some naturally bounded specific characteristic features of some microorganisms tend to resist or prevent from the toxins and even kill them.It is just coming from such actual need that we have found that the element Agis endowed with the instinct to induce the toxicity even at lower levels（0.01 mmol/L）,while K.pneumonia have a variable tolerance for Pb and Cr at 1.0 mmol/L and 5.0mmol/L,correspondingly.It is also coming from this need that we have investigated and identified some protein expression profiles of K.pneumonia via the two-dimensional electrophoresis associated with the mass spectrometry.As compared with the intact group and all of the significant spots,we have found that there ex-ist approximately 458,609 and 558 visualized protein spots on the 2-DE gel reliably in all the biological replicates on the condition when the cells were exposed to Ag^＋,Pb^2 ＋and Cr^3 ＋for 20 hours,respectively.As for the differentially expressed proteins,though there may exist great difference in estimation of the quantity and the expression level of each group,it is an undeniable fact that there exist at least thirteen kinds of proteins with the common changes for all the three groups,including the three open-up proteins and the ten close-down ones.Thus,in accordance with the gene ontology,it would be possible to divide the thirteen proteins into six types,that is,the intramolecular transfer,transport,oxidoreduction,transcription,translation,energy production and conservation.Since heavy metals tend to disturb the cell redox systems,they can make the greatest contribution to the toxicity by generating the reactive oxygen（ROS） and nitrogen（RNS）
