肾阳虚致高脂血症大鼠中FAS蛋白对甘油三酯的调控作用及机制研究  被引量:4

摘  要:目的:通过肾阳虚制作高脂血症模型,观察右归丸对模型大鼠FAS蛋白及其上游LXRα等因子表达的影响,以探讨肾阳虚对模型大鼠甘油三酯(TG)的影响机制。方法:利用大剂量肌注氢化可的松制作肾阳虚高脂血症大鼠模型,给予右归丸进行干预,通过蛋白免疫印迹法(Western blot)检测实验大鼠肝组织FAS蛋白、LXRα和SREBP-1c表达,通过免疫组化检测实验大鼠肝组织FAS蛋白的表达。结果:大剂量肌注氢化可的松成功复制出了肾阳虚高脂血症的模型大鼠,与正常组比较,右归丸组和模型组肾阳虚高脂血症大鼠肝内LXRα、SREBP-1c和FAS蛋白表达增加(P<0.05),与模型组比较,右归丸组大鼠肝内LXRα、SREBP-1c和FAS蛋白表达减少(P<0.05);结论:肌注氢化可的松制造的肾阳虚高脂血症模型可能是上调了LXRα、SREBP-1c和FAS的蛋白的表达而影响了血液中TG的代谢,右归丸可能通过抑制LXRα、SREBP-1c和FAS蛋白的表达来降低模型大鼠TG的含量。Objective: Through the kidney yang deficiency making hyperlipidemia model, to observe the effect of You- gui Pill on the expression of Fas protein in model rats and its upstream LXR alpha factor and explore the mechanism of the kidney yang deficiency model rats' triglyceride (TG). Methods: Kidney yang deficiency high - fat hyperlipidemia rat model was made by intramuscular injection of large dosage of hydrocortisone and the models were given Yougui Pill inter- vention. By protein immunoblotting (Western blot) assay, Fas protein, LXt/and SREBP expressions in liver were detec- ted and lay inmmnohistochemistry experiments the expression of Fas protein in rat liver tissues was detected. Results: Large dose of intramuscular injection of hydroeortisone successfully replicated the kidney yang deficiency of hyperlipi- demia model rats. Compared with the normal group, Yougui Pill group and model group' s hepatic LXR and SREBP LC and Fas protein expression were increased (P 〈 0. 05 ) and compared with model group, Yougui Pill group' s hepatic LXR and SREBP LC and Fas protein expressions were decreased (P 〈 0. 05 ). Conclusion: Intramuscular injection of hydro- cortisone induced hyperlipidemia model may raise the LXtl alpha and SREBP - I e and Fas protein expressions and influ- ence the blood TG metabolism. Yougui Pill may through inhibiting LXR alpha and StlEBP - 1 c and Fas protein expres- sion in rats to lower TG.
