摘 要:目的:观察炮制前后白术醇提取物对豚鼠离体肠肌的作用,评价麸炒炮制工艺对白术药效的影响。方法:采用豚鼠离体肠肌实验,经张力换能器和生物信号采集系统,用炮制前后的白术醇提取物,分别刺激离体肠肌和经1.0μg/m L乙酰胆碱预处理的离体肠肌,测量肠肌肌张力变化,观察炮制前后白术对离体肠肌张力及幅度的变化,并与阿托品的作用进行比较。结果:生白术醇提物及麸炒白术醇提物在0.2~4.0 mg/m L浓度范围内对离体肠肌具有抑制作用,4.0 mg/m L时抑制作用最大,其抑制作用与4.0μg/m L的阿托品相当,且麸炒后抑制作用增强,抑制率为76.5%;生白术醇提物及麸炒白术醇提物对乙酰胆碱所致的肠肌兴奋具有显著的抑制作用,给药浓度为2.0 mg/m L时作用最强,麸炒后作用增强,抑制率为49.3%。结论:白术醇提物对肠肌具有抑制作用,麸炒炮制后抑制作用增强。Objective : To observe the effect on isolated intestinal muscle of guinea pig with the methanol extract of atraetylodes before and alter processing and evaluate the ettects of atractylodes with different processing technology. Methods : The intestinal muscle of guinea pig was isolated. The isolated muscle and the prelreated muscle with 1.0 μg/ mL aeetyleholine were stinmulated using the methanol extracts of Atractylodis. respectively. The tension and range of muscle contraction were measured with tension transducer and biological signal acquisition system. The changes were ob- stowed to evaluate the influence of processing on the effects of Atractylodis. The effects of Atractvlndes before and after processing were cmnpared with atropine. Result: The alcohol exlracts of atractylodes befiwe and after processing were ca- pability for inhibiting the isolated intestinal muscle contraction in the range of 0.2 - 4.0 mg/mL. The inhibition was the most in the dose of 4.0 mg/mL, and it was ahnost the same effect of atropine in 4.0 μg/mL. And inhibition was enhanced after processing, the inhibition rate was 76.5%. There was a significant inhibitory effect on the mHscle pretreated with aeetyleholine both using the methanol extract of atraetylodes before and after processing. The inhibition was the most in the dose of 2.0 mg/mL and inhibition was enhanced after processing. The inhibition rate was 49.3%. Conclusion: The methanol extract of Atractylodis can inhibit the isolated intestinal nmscle conh'action and the effect was enhanced after processing.
