清热益气中药对自发性2型糖尿病大鼠胰岛细胞凋亡与炎症因子表达的影响

摘　　要：目的:研究清热益气中药黄连人参对药对自发性2型糖尿病大鼠胰岛细胞凋亡与炎症因子表达的影响。方法:采用高热量高脂饲料喂养诱导GK大鼠,建立自发性T2DM大鼠模型,予清热益气中药黄连人参对药进行干预4周,设Wistar大鼠作为对照,观察超敏C反应蛋白质、肿瘤坏死因子α水平及凋亡因子Bcl-2,Bax的表达情况。结果:模型组血清TNF-α显著高于正常组和治疗组(P<0.01),模型组和治疗组血清hs-CRP水平均显著高于正常组(P<0.01),模型组TNF-α在胰岛的表达水平显著高于正常组和治疗组(P<0.01,P<0.05),模型组Bax表达水平显著高于正常组和治疗组(P<0.01)。结论:清热益气中药黄连人参可降低糖尿病大鼠炎症因子水平,下调炎症因子及促凋亡因子的表达,抑制细胞凋亡,促进胰岛细胞损伤修复,也提示了2型糖尿病"壮火食气"的微观机制。Objective ： To study the effect of heat - clearing qi - replenishing herb berberine and ginseng on pancreas B - cell apoptosis and expression of inflammatory cytokines in spontaneous type 2 diabetic rats. Methods ： The spontaneous T2DM rat model was established with the high calorie high fat diet - induced GK rats. And then they were divided into the model group and treatment group that were intervened with berberine and ginseng for four weeks. Meanwhile, Wistar rats were set as a blank control group. We observed the levels of hypersensitive C - reactive protein, tumor necrosis factor - α and the expressions and apoptosis factor Bel - 2, Bax in the pancreas as well. Results ： Serum levels of TNF - α of the model group were significantly higher than those of the normal and treatment group （P 〈 0. 01 ）. Serum levels of hs - CRP of the model and treatment group were significantly higher than those of the normal group （P 〈 0. 01 ）. The expres- sion level of TNF - ct in pancreatic islets of the model group was significantly higher than that of the other two （ P 〈 0. 01, P 〈0. 05）. The expression level of Bax of model group was significantly higher than others （P 〈0. 01 ） as well. Conclu- sion ： Heat - clearing and qi - replenishing herb berberine and ginseng can reduce the levels of inflammatory factors in di- abetic rats and reduce the expressions of inflammatory cytokines and apoptotic factors, inhibit the process of apoptosis and promote islet cell repairmen. It also suggests the micro - mechanism of TCM huge fire consuming qi mechanism of type 2 diabetes.
