摘 要:目的:探讨欢乐宁方对围绝经期综合征小鼠下丘脑KNDy编码基因及其相关受体信号通路的影响。方法:建立11月龄ICR小鼠围绝经期综合征模型,随机分为模型组、欢乐宁方低、中、高剂量组、补佳乐组,另取2月龄小鼠作为正常组,每组各10只;测定各组小鼠血清E_2、LH、FSH水平,检测下丘脑组织Ki SS-1、NKB、DYN及相关受体GPR54、NK3和KOR mRNA的表达。结果:围绝经期小鼠血清E2明显降低,血清FSH、LH明显升高;Kiss-1、GPR54、NKB、NK3、DYN、KOR mRNA表达量明显上升。欢乐宁方能明显升高围绝经期小鼠血清E2水平,降低血清FSH、LH水平,能下降下丘脑Kiss-1、GPR54、NKB、NK3、DYN、KOR mRNA的表达,以中高剂量为佳。结论:欢乐宁方能有效调节围绝经期综合征小鼠的性激素水平;欢乐宁方可能通过作用于KNDy神经元,从而影响GnRH的分泌来调节H-P-O轴,这可能是欢乐宁方治疗围绝经期综合征的作用机制之一。Objective : To explore the effet of the signaling pathway changes of Kisspeptin, neurokinin B and dynorphin (KNDy) and their related receptors GPR54, NK3 and KOR in hypothalanms of perimenopausal mice by Huanlening Recipe. Methods: Fifty 11 -month -old ICR mice as perimenopausal mice were randomly divided into model group, Huanlening Reeipe low, middle and high dose groups, and progynova group, n = 10. And 10 another 2 - month - old ICR mice were used as the normal group. At the end of the experiment,measure the serum E2, LH and FSH levels and detect the expressions o1' KiSS - 1, NKB and DYN and related reeeptors GPR54, NK3 and KOR mRNA in hypothalamus with Real -time quantitative PCR. Results: 3"he levels of" serum E2 of perimenopausal mice were decreased significantly while serum FSH and LH were increased significantly. The expressions of Kiss - 1, GPR54, NKB, NK3, DYN and KOR mRNA were increased signifieantly. Huanlening Recipe signifieantly increased the serum E2 levels of perimenopausal mice while decreased the serum FSH and LH levels and decreased the expressions of Kiss - 1 , GPR54, NKB, NK3, DYN and KOR mRNA in hypothalamus, and the eftect of middle and high dose of Huanlening Recipe was better. Conclusion:Huanlening Recipe can regulate the sex hormone levels of perimenopausal mice effectively and may affect the secretion of GnRH to regulate H - P - 0 axis by acting on the KNDy, which may be one of the meehanisms of treating perimenopausal syndrome of Huanlening Recipe.
