摘  要:目的评价七氟醚对大鼠海马神经元d7烟碱型乙胆碱受体(α7nAChR)表达的影响。方法清洁级健康成年sD大鼠48只,体重210~280g,3—4月龄,雌雄不拘,采用随机数字表法分为4组(n=12):对照组(C组)和1%、3%和5%七氟醚组(S1组、S2组和S3组),S1组、S2组和S3组分别吸入相应浓度七氟醚1h。分别于麻醉后1和7d时行水迷宫实验,评价认知功能。分别于水迷宫实验结束后处死6只大鼠取海马,采用Westernblot法检测α7nAChR表达,ELISA法检测TNF—α和IL-1的含量。结果与C组比较,S1组麻醉后1和7d时穿越平台次数减少,S2组和S3组麻醉后1和7d时目标象限停留时间缩短,穿越平台次数减少,原平台所在象限停留时间比率和原平台所在象限游泳路程比率降低,S1组、S2组和S3组麻醉后1和7d时海马TNF—α和IL-1的含量升高,海马α7nAChRs表达下调(P〈0.05);与S1组比较,S2组和S3组麻醉后1和7d时海马α7nAChRs表达下调(P〈0.05);与S2组比较,S3组麻醉后1d时海马α7nAChRs表达下调(P〈0.05)。S1组、S2组和S3组间水迷宫实验各指标比较差异无统计学意义(P〉0.05)。结论七氟醚诱发海马炎症反应的机制与下调海马神经元α7nAChR表达有关。Objective To evaluate the effect of sevoflurane on the expression of a7 nicotinic acetylcholine receptor ( α7nAChR ) in hippocampal neurons of rats. Methods Forty-eight pathogen-free healthy adult Sprague-Dawley rats of both sexes, weighing 210-280 g, aged 3-4 months, were divided in- to 4 groups (n= 12 each) using a random number table: control group (group C) and 1%, 3% and 5% sevoflurane groups (S1, S2 and S3 groups). In S1, S2 and S3 groups, 1%, 3% and 5% sevoflurane were inhaled for 1 h, respectively. Morris water maze test was performed at 1 and 7 days after anesthesia to assess the cognitive function. Six rats in each group were sacrificed after the end of the test. The brains were then removed and the hippocampi were isolated for detection of the expression of α7nAChR ( by Western blot) and contents of tumor necrosis factor-alpha and interleukin-1 ( by enzyme-linked immunosorbent as- say). Results Compared with group C, the number of crossing the platform was significantly decreased at 1 and 7 days after anesthesia in group S~, the time of staying at the target quadrant was significantly short- ened, the number of crossing the platform was decreased, and the rate of time of staying at the original platform quadrant and rate of swimming distance at the original platform quadrant were decreased at 1 and 7 days after anesthesia in Sz and S3 groups, and the contents of tumor necrosis factor-alpha and interleukin-1 were significantly increased, and the expression of c^7nAChR was down-regulated at 1 and 7 days after anes- thesia in S~, S2 and S3 groups (P〈0.05). Compared with group S~, the expression of c^7nAChR was sig- nificantly down-regulated at 1 and 7 days after anesthesia in $2 and S3 groups (P〈0. 05). Compared with group S2 , the expression of c^TnAChR was significantly down-regulated at 1 day after anesthesia in group S3 (P〈0. 05). There was no significant difference in each parameter of Morris water maze test among group S~, group S2 and group S3 (P〉0.
