摘  要:目的评价异氟醚预处理对局灶性脑缺血再灌注大鼠海马含GluR1亚基α-氨基-3-羟基-5-甲基-4-异恶唑丙酸(AMPA)受体表达的影响。方法清洁级健康雄性SD大鼠48只,体重250~280g,7~8周龄,采用随机数字表法分为3组(n=16):假手术组(S组)、局灶性脑缺血再灌注组(I/R组)和异氟醚预处理组(IPC组)。采用线栓法阻塞大脑中动脉2h后恢复灌注的方法制备大鼠局灶性脑缺血再灌注诱发大鼠认知功能降低模型。IPC组每天吸入1.5%异氟醚1h,连续5d,末次预处理后24h制备模型。于术后9和23d时行水迷宫实验,连续6d;于术后14和28d时处死大鼠取海马,采用RT—PCR法检测GluR1mRNA表达。采用Westernblot法检测GluR1表达。结果与s组比较,I/R组与IPC组术后各时点逃避潜伏期延长,穿越平台次数、原平台象限游泳距离比减少,术后14d时海马GIuR1及其mRNA表达下调(P〈0.05);与I/R组比较,IPC组术后10~13d时逃避潜伏期缩短,术后14d时穿越平台次数、原平台象限游泳距离比增多,海马GluR1及其mRNA表达上调(P〈0.05)。结论异氟醚预处理改善局灶性脑缺血再灌注大鼠认知功能的机制与其上调海马含GluR1亚基AMPA受体表达有关。Objective To evaluate the effect of isoflurane preconditioning on the expression of hippocampal GluR1 subunits-containing α-amino-3-hydroxy-5-methyl-4-isoxazolepropionate (AMPA) receptors in a rat model of focal cerebral ischemia-reperfusion (I/R). Methods Forty-eight pathogen-free healthy male Sprague-Dawley rats, weighing 250-280 g, aged 7-8 weeks, were divided into 3 groups (n= 16 each) using a random number table: sham operation group (group S) , focal cerebral [/R group (group I/R) and isoflurane preconditioning group (group IPC). Focal cerebral I/R was induced by occlu- sion of the middle cerebral artery for 2 h followed by reperfusion to induce cognitive decline in I/R and IPC groups. Rats were exposed to 1.5% isoflurane for 1 h every day for 5 consecutive days, and the model was established at 24 h after the last exposure in group IPC. Eight rats in each group were selected to perform Morris water maze test for 6 consecutive days starting from 9 or 23 days after operation. The rats were sacri- ficed at 14 and 28 days after operation, and the hippocampal tissues were obtained for determination of the expression of GluR1 mRNA ( by using real-time polymerase chain reaction) and GluR1 protein (by Western blot). Results Compared with group S, the escape latency was significantly prolonged at each time point after operation, the frequency of crossing the original platform quadrant and percentage of swimming distance at the original platform quadrant were decreased at 14 and 28 days after operation, and the expression of GluR1 protein and mRNA was down-regulated at 14 days after operation in I/R and IPC groups (P〈 0.05). Compared with group I/R, the escape latency was significantly shortened at 10-13 days after opera- tion, the percentage of swimming distance at the original platform quadrant and frequency of crossing the o- riginal platform quadrant were increased at 14 days after the operation, and the expression of GluR1 protein and mRNA was up-regulated at 14 days aft
