摘  要:目的评价黄芪甲苷对老龄大鼠术后认知功能的影响。方法健康雄性SD大鼠60只,22月龄,体重360~480g,采用随机数字表法,将其分为4组(n=15):对照组(C组)、手术组(S组)、低剂量黄芪甲苷组(L—AGS组)和高剂量黄芪甲苷组(H—AGS组)。术前3d,L-AGS组和H.AGS组分别腹腔注射黄芪甲苷20或40mg/kg,1次,d,C组与S组腹腔注射等容量生理盐水。吸入1-8%异氟醚麻醉下行脾切除术。于术后1d时随机处死5只大鼠,取海马组织,采用ELISA法测定海马IL-1β、TNF—α和1L-6含量;采用Westernblot法测定海马活化caspase-3、Bax和Bcl-2的表达水平。剩余大鼠于术后15d时行Morris水迷宫实验。结果与c组比较,s组术后逃避潜伏期延长,穿越平台次数减少,探索时间缩短,海马活化caspase-3和Bax表达上调,Bcl-2表达下凋,IL-1B、TNF—α和IL-6含量升高(P〈0.05);与s组比较,L—AGS组和H—AGS组术后逃避潜伏期缩短,穿越平台次数增加,探索时间延长,海马活化caspase-3和Bax表达下调,Bcl-2表达上调,IL-1β、TNF—α和IL-6含量降低(P〈0.05);与L—AGS组比较,H—AGS组术后逃避潜伏期缩短,穿越平台次数增加,探索时间延长,海马TNF—α含量降低(P〈0.05)。结论黄芪甲苷可改善老龄大鼠术后认知功能,且与剂量有关。Objective To evaluate the effect of astragaloside IV on postoperative cognitive function in aged rats. Methods Sixty healthy male Sprague-Dawley rats, aged 22 months, weighing 360-480 g, were divided into 4 groups (n= 15 each) using a random number table: control group (group C), surgery group (group S) , low-dose astragaloside Ⅳ group (group L-AGS) and high-dose astragaloside IV group (group H-AGS). At 3 days prior to surgery, astragaloside IV 20 and 40 mg/kg were injected intraperitoneally once a day in L-AGS and H-AGS groups, respectively. The equal volume of normal saline was given instead in C and S groups. The animals underwent splenectomy under anesthesia with 1.8% isoflurane in S, L-AGS and H-AGS groups. Five rats in each group were randomly sacrificed at 1 day after operation, the hippocampi were removed for determination of interleukin-lbeta (IL-113) , tumor necrosis factor-alpha (TNF-a) and IL-6 contents (by enzyme-linked immunosorbent assay ) and expression of activated caspase-3, Bax and Bcl-2 (by Western blot). The left animals underwent Morris water maze test at 15 days after operation. Results Compared with group C, the escape latency was significantly prolonged, the frequency of crossing the original platform was reduced, the space exploration time was shortened, the ex- pression of activated caspase-3 and Bax was up-regulated, the expression of Bcl-2 was down-regulated, and the IL-1β, TNF-α and IL-6 contents were increased after operation in group S (P〈0.05). Compared with group S, the escape latency was significantly shortened, the frequency of crossing the original platform was increased, the space exploration time was prolonged, the expression of activated caspase-3 and Bax was down-regulated, the expression of Bcl-2 was up-regulated, and the IL-113, TNF-α and IL-6 contents were decreased after operation in L-AGS and H-AGS groups (P〈0.05).
