摘  要:目的:研究神经干细胞(Neural stem cells,NSCs)植入皮肤全层缺失后对创面的影响。方法:分离并培养SD大鼠骨髓间质干细胞(bone marrow mesenchymal stem cells,BMSCs),将酸性成纤维细胞生长因子(Acidic fibroblast growth factor,a FGF)与DMEM/F12配成最佳诱导浓度80 ng/m L,将BMSCs诱导分化为神经干细胞,DAPI标记。根据实验设计,将54只SD大鼠背部脊柱两侧造模,形成皮肤软组织创面。按随机分为局部注射NSCs(A组)、局部创面注射BMSCs(B组)、局部创面注射等量生理盐水(C组);术后3、7、14 d,分别观察记录创面愈合时间,计算创面的愈合率,同时切取标本,石蜡包埋处理切片,标本组织行HE染色和VG染色反映皮肤创面炎症反应及组织胶原纤维的增生情况。结果:NSCs局部注射移植后可促进神经修复,在实验过程中观察NSCs对创面愈合的影响,A组创面愈合时间短较B、C组明显缩短(F=10.360,P=0.000));痂皮薄,创面稍燥,HE染色镜下观察局部炎症反应轻微,无出血、积脓、坏死;A组愈合率明显高于B组阳性对照组,B组阳性对照组高于生理盐水阴性对照组(F=1.323,P=0.000)。结论:神经干细胞作为种子细胞移植局部皮肤创面后,可修复创面神经,神经分泌的各种神经肽类物质可加速皮肤创面的修复,缩短修复时间,减轻瘢痕。提高患者满意度。Objective: To observe the effects of neural stem cells (NSCs) on wound healing of full-thickness skin lesion in rats. Methods: Rat bone marrow mesenchymal stem cells(BMSCs) were isolated and cultured in DMEM/F12 supplemented with Acidic fibroblast growth factor (Acidic fibroblast growth factor, aFGF) at 80ng/ml to induce the differentiation of BMSCs into neural stem cells that were then labeled with DAPI. Skin lesion model was created using the method described in materials and methods on the back of SD rats. A total of 54 model rats were randomly divided into the local wound injection group (NSCs, group A), local BMSCs injection group (group B), and local normal saline injection group (group C). Wound healing time and the rate of wound healing were recorded on day 3, 7 and 14 postoperation,and at the same time tissues were collected, fixed, paraffin embedded, and stained using HE and VG to evaluate the inflammation and fibrosis. Results: After the artificial skin transplantation, the wounds in group A healed well , accompanied by slight local inflammatory response without bleeding, empyema and necrosis, and the wound healing time in group A was significantly shortened compared with group B and group C(P = 0.000). The healing rate in NSCs group was the highest, followed by BMSCs group and saline control group (P = 0.000 in both). Conclusion: Neural stem cells transplanted into skin wounds by local injection can be used to repair skin wounds.
