摘　　要：目的探讨在重度子痫前期患者胎盘组织中转录因子激活蛋白-2α(AP-2α)、B淋巴细胞瘤-2基因(Bcl-2)及原癌基因(C-myc)的表达情况及临床意义。方法选取86例于2013年5月至2014年5月期间行剖宫产术分娩的产妇,分为正常对照组(n=43)与重度子痫前期组(n=43),应用实时荧光定量PCR(RT-PCR)和免疫组化法检测两组产妇胎盘组织中AP-2α、Bcl-2和C-myc的表达。结果重度子痫前期组胎盘组织中AP-2α蛋白阳性表达显著高于对照组(P<0.05),C-myc、Bcl-2蛋白阳性表达均显著低于正常对照组(P<0.05);相较于对照组,重度子痫前期组中AP-2αmRNA表达明显升高,Bcl-2 mRNA和C-myc mRNA表达明显降低(P<0.05)。结论在重度子痫前期胎盘组织中C-myc、Bcl-2的表达下降,AP-2α表达上升,这种差异表达可能参与疾病的病理生理过程。Objective To explore the clinical significance of expression of transcription factor activated protein-2 alpha（ AP-2α）,B lymphocyte tumor2 gene（ Bcl-2） and C-myc in placenta tissue of patients with severe preeclampsia. Methods 86 pregnant women with cesarean in this hospital during May 2013 to May 2014 were selected for this study,and they were divided into normal control group（ n = 43） and severe preeclampsia group（ n = 43）. The expression levels of of AP-2α,Bcl-2 and C-myc in patients of these two groups were detected by real time fluorecence quantitative PCR（ RT-PCR） and immunochemical method. Results The positive expression of AP-2α protein in placenta tissue of patients in severe preeclampsia group was significantly higher than that in patients of control group（ P〈0. 05）. The expression levels of C-myc and Bcl-2 protein were significantly lower than those of normal control group（ P〈0. 05）. In comparison with the control group,the expression of AP-2α mRNA in severe preeclampsia group was significantly higher,and the expression of C-myc mRNA and Bcl-2m RNA was significantly lower（ P〈0. 05）. Conclusion The expression levels of of Bcl-2 and C-myc are decreased and eexpresssion level of AP-2 α is increased in placenta tissue of patients in severe preeclampsia group,these may be involved in the pathophysiological process of the disease.
