摘 要:目的:探讨利拉鲁肽对2型糖尿病(type 2diabetes mellitus,T2DM)大鼠体脂分布和骨密度的影响。方法:高脂饮食联合2%链脲佐菌素诱导T2DM模型,皮下注射0.1 mg/kg(小剂量组)和0.2mg/kg(大剂量组)利拉鲁肽干预4周。DM模型组不干预。另设正常饮食组和高脂喂养的单纯肥胖组为对照。动态观察大鼠体重变化。4周末应用核磁共振成像观测全身体脂分布,骨密度仪检测骨密度值,腹主动脉取血测定血液生化学指标。结果:与DM组比较,利拉鲁肽大、小剂量组体重明显减低,血脂和糖化血红蛋白下降,大剂量组更明显(P<0.05)。单纯肥胖组和DM组体脂分布最高,利拉鲁肽干预后全身体脂明显减少。利拉鲁肽组骨密度介于正常饮食组和DM组之间(P<0.05)。结论:利拉鲁肽可有效减轻糖尿病大鼠体重,降低血脂,调节全身体脂分布,提高骨密度。ObjectiveTb investigate the effect of liraglutide on body fat tissue and bone mineral density (BMD) in rats with type 2diabetes mellitus (T2DM ). Methods: Tlie T2DM rat model was established by feeding high-fat diet for 12 weeks and followed byintraperitoneal injection of streptozotocin (STZ). Then the rats were administered by subcutaneous injection twice daily in a dose of 0.1 mg/kg (low-dase group) or 0.2 mg/kg (high-dose group) liraglutide for 4 weeks. The rats in normal diet group were given ordinary diet and therats in obese group were accepted liiglvfat diet. The body weight was observed dynamically. At the end of experiment?bony marrow density(BMD) was dctcctcxl by X-ray osteodensitometer,and the body fat distribution was measured by MRI,and the scrum biochemical parameterswere determined. Results :Cx〉mp?ired with DM group? liraglutide administnition ob/a〇usly reduced Ixxly weight index of dialx^tic rats (P〈C0.05), and the serum lipid and glycosylated hemoglobin level were also significantly decreased (P〈C〇. 05), which was more prominent inhigh-dose liraglutide group ( P 〈 0 .05). MRI showed the total body fat distribution was the highest in obese group and DM group, andliraglutide intervention remarkably diminishcxl the body fat. The BMD value in liraglutide group was higher th^in DM group? but less thannormal diet groupCP^O. 05). Conclusion:Liraglutide treatment can effectively reduce txxly weight,decrease serum lipid,adjast fat distributionand increase BMD in type 2 diabetic rats.
