摘  要:目的建立HPLC法测定行连续性肾脏替代治疗患者肾脏超滤液中利奈唑胺的含量,反应滤器对利奈唑胺的清除情况,为其给药剂量调整提供可靠依据。方法选用Agilent ZORBAX SB-C_(18)(4.6mm×150mm,5μm)色谱柱;流动相为甲醇-水(45∶55),流速为1.0ml·min^(-1);检测波长:254nm;柱温:25℃;进样量:10μl;运行时间6min。采集行连续性肾脏替代治疗重症医学科患者肾脏超滤液,-20℃冻存,经冷冻干燥处理后,复溶进行含量测定。结果利奈唑胺在1.00~40.00μg·ml-1范围内与其峰面积线性关系良好(r=0.9999),平均回收率为99.54%(n=9),RSD为0.87%。结论本方法简单、快速,结果准确、重复性好,适用于行连续性肾脏替代治疗肾脏超滤液中利奈唑胺的含量测定,能够反应滤器对利奈唑胺的清除情况。Objective To establish an HPLC method for the determination of linezolid in continuous renal replacement therapy(CRRT)ultra-filtrate liquid for patients in intensive care units,to present its clearance by the filter and to provide data for dosage adjustment.Methods Agilent ZORBAX SB-C18(4.6mm×150mm,5μm)column was chosen for analysis.The mobile phase consisted of methanol and water(45∶55).The flow rate was 1.0 ml·min^-1 at a detection wavelength of 254 nm.The temperature was set at 25 ℃,the injection volume was 10μl,and the running time was 6 min.Samples were collected from an intensive care unit,frozen at-20 ℃,freeze dried and determined by HPLC.Results A good linear relationship of fluconazole was observed within the range of 1.00-40.00μg·ml^-1(r=0.9999).The average recovery was 99.54%(n=9),with an RSD of 0.87%.Conclusion This method is quick,simple,accurate,robust and suitable for determination of fluconazole concentration in CRRT ultra-filtrate liquid.
