摘  要:目的观察姜黄素对Lewis肺癌裸鼠Bcl-2、Bax和p53蛋白表达的影响。方法BALB/C裸鼠平均分为3组:空白组、模型组和姜黄素组,每组10只。各组小鼠右腋皮下接种Lewis瘤细胞,空白组除外。空白组与模型组灌胃0.5%的羟甲基纤维素钠溶液,姜黄素组灌胃180 mg/kg姜黄素,连续7 d,每日1次。于末次给药后处死小鼠,计算抑瘤率,采用Western blotting法检测Bcl-2、Bax和p53蛋白的表达。结果经姜黄素干预后,Lewis肺癌小鼠的肿瘤体积显著缩小,Bcl-2和p53蛋白表达显著减少,Bax的表达显著升高。结论姜黄素对lewis肺癌肿瘤生长有抑制作用,其机制可能和下调Bcl-2和P53水平,上调Bax水平有关。Objective: To investigate the effects of curcumin on Bcl-2,Bax and p53 expression in Lewis lung nude mice. Methods: Nude mice were divided into three groups: control group,tumor model group and cureumin treatment group (180 mg/kg) with 10 ,,lice in each group. For preparation of transplanted tumor model, Lewis cells were subcutaneously injected into the right limb armpit of the BALB/C nude mice,except the control group. The mice of control group and tumor model group received 0.5% CMC-Na solution by intragastric administration, once a day for 7 days. The mice of curcumin treatment group received suspension of eurcumin 180 mg/kg by garage once a day for 7 days. After final administration, the mice were killed and tumor inhibition rate was measured. Bcl-2,Bax and p53 protein expression was analyzed by Western blotting. Results: The lung tumor size reduced significantly in curcumin group. Bcl-2 and p53 protein expression decreased significantly in lung tumor tissues, while Bax expression increased. Conclusion: Curcumin inhibits the development of lung tumor through downregulating Bcl-2 and p53 expression and up-regulating Bax expression.
