接骨续筋口服液对家兔骨折愈合过程BMP-2、TGF-β1表达的影响

摘　　要：目的:观察接骨续筋口服液主要成分对兔骨折愈合过程中骨痂中BMP-2、TGF-β1的影响。方法:选取健康8个月龄长耳新西兰大白兔80只,随机取12只作为假手术组;其余60只制作骨折模型后再分为5组,分别为模型组、对照组、接骨续筋口服液低、中、高剂量组,每组12只;余8只预留意外死亡。于术后第2天开始给药,假手术组及模型组动物灌等容量的蒸馏水;接骨续筋口服液按低剂量组、中剂量组、高剂量组灌胃给药;对照组以云南白药灌胃给药。每组分批于术后1、2、4周各处死3只,以检测接骨续筋口服液对不同时期骨折愈合过程中骨痂中BMP-2、TGF-β1表达影响。结果:在家兔骨折造模后愈合中1W,2W,4W接骨续筋口服液高剂量组骨痂中BMP-2、TGF-β1明显高于其余各组。结论:接骨续筋口服液能通过提高骨折后骨痂内BMP-2、TGF-β1的表达,从而促进骨折愈合。Objective： To observe the influence of the main ingredients in the Jiegu Xujin oral solution to the TGF- β1 and the BMP-2 in callus during the rabbits＇ fracture healing process. Methods： 80 healthy New Zealand long-eared white rabbits with 8 months, 12 rabbits were chose as control group randomly. The other 60 rabbits divided into five groups after making fracture model, which is model group, control group, low dose group, medium dose groups and high dose groups, 12 rabbits in each group. The remaining eight rabbits were death by accident. The rats in the control group and the model group were given the same volume of distilled water. The rats were treated with Jiegu Xu- jin oral solution according to low dose, middle dose and high dose. The control group was treated with Yunnan Baiyao intragastric administration. Three rats in each group was sacrificed at 1, 2 and 4 weeks after operation to detect the ef- fect of Jiegu Xujin oral solution on the expression of BMP-2 and TGF- 13 1 in callus during different periods of fracture healing. Results： The levels of BMP-2 and TGF- β 1 in the callus of 1W, 2W and 4W in the high dose group were sig- nificantly higher than those in the other groups after fracture healing. Conclusion： Jiegu Xujin oral solution can improve the expression of BMP-2 and TGF- β 1 in the callus after fracture.
