灌肠散对大鼠溃疡性结肠炎的治疗作用研究

摘　　要：目的探讨灌肠散对溃疡性结肠炎大鼠的治疗作用并初步探索其作用机制。方法雄性SD大鼠随机分为6组:正常对照组,模型组,灌肠散低、中、高剂量组及柳氮磺吡啶组,每组10只。使用2,4,6-三硝基苯磺酸造模后,保留灌肠给药7 d,正常对照组及模型组给予生理盐水。采用ELISA法测定血清中IL-6、IL-8、IL^(-1)0及TNF-α含量,取结肠,称取结肠湿重,并进行DAI、CMDI和HS评分。结果与正常组比较,除IL^(-1)0显著降低(P<0.01)外,模型组大鼠各项指标均显著升高(P<0.01)。与模型组比较,除IL^(-1)0显著升高(P<0.01)外,灌肠散低、中、高剂量组及柳氮磺吡啶组大鼠各项指标均显著降低(P<0.01)。结论灌肠散对TNBS-乙醇复制的溃疡性结肠炎大鼠模型有治疗效果;可能通过降低促炎细胞因子IL-6、IL-8、TNF-α表达,促进抗炎细胞因子IL^(-1)0表达,平衡溃疡性结肠炎大鼠血清中细胞因子水平,从而对溃疡性结肠炎起到治疗作用。Objective To explore the therapeutic effect of Guanchang powder on ulcerative colitis （UC） in rats and explore its mechanism. Methods Male sprague-dawley （SD） rats were randomly divided into 6 groups： a normal control group, a model group, Guanchang powder low, medium and high dose groups and sulfasalazine （SASP） group with 10 rats in each group. The UC rats were induced by intracolonic administration of 2, 4, 6-trinitrobenzene sulfonicacid. After that, Guanchang powder and SASP were given by retention enema daily for 7 days, and saline for the normal control group and the model group. The plasma levels of IL-6, IL-8, IL- 10, and TNF-α were measured by ELISA. The colonic wet weight was obtained and assessed with DAI, CMDI and HS. Results Compared with the normal group, all the indexes in the model group were significantly increased （P 〈 0.01） except for the significant reduction of IL-10 （P 〈 0.01）. Compared with the model group, all the indexes in the Guanchang powder low, medium, high dose groups and SASP group were significantly reduced （P 〈 0.01） except for the significant increase of IL-10 （P 〈 0.01）. Conclusion Guanchang powder has therapeutic effect on TNBS and ethanol replication of UC rat models. It can decrease the expression of proinflammatory cytokines IL-6, IL-8, and TNF-α, promote the expression of anti-inflammatory cytokine IL-10 and balance serum cytokine levels in UC rats, thus treating UC.
