摘 要:本试验旨在研究饲粮铁水平对肉仔鸡锰吸收及组织中沉积的影响及其机制。将336只1日龄商品代罗斯308肉公雏按体重随机分为4个组,每组6个重复,每个重复14只鸡,分别饲喂不加铁基础饲粮(含铁77.7 mg/kg)、以硫酸亚铁(Fe SO4·7H2O)形式添加铁100、250、500 mg/kg饲粮。28日龄进行原位结扎十二指肠灌注8.74 mmol/L锰测定锰吸收率。结果表明:血浆铁和血浆转铁蛋白饱和度均随饲粮铁水平升高而提高(P<0.10);胰腺、十二指肠黏膜和胫骨灰铁含量随饲粮铁水平升高而升高(P<0.10),锰含量随饲粮铁水平升高而降低(P<0.10);3个加铁组十二指肠黏膜二价金属转运蛋白m RNA水平均显著低于对照组(P<0.10),添加铁500 mg/kg组膜转铁蛋白m RNA水平显著低于对照组和其他2个加铁组(P<0.10);加铁组十二指肠锰吸收率均显著低于对照组(P<0.10)。结果提示,饲粮铁可能通过调控十二指肠黏膜二价金属转运载体的表达降低锰的吸收及组织中的沉积。Abstract: This experiment was conducted to investigate the effect of dietary iron (Fe) level on manganese (Mn) absorption and deposition in broilers. A total of 336 1-day-old Rose-308 male chicks were divided into 4 groups with 6 replicates and 14 chicks each replicate cage, and fed the basal diet (control, containing Fe 77.7 mg/kg) or basal diet supplemented with 100, 250, or 500 mg Fe/kg for 28 days. Ligated duodenal loops were perfused with 8.74 mmol/L of Mn to determine the Mn absorption. The results showed that plasma Fe content and transferrin saturation increased (P〈0.10) as dietary Fe level increased. The Fe contents in pancreas, duodenal mucosa, and tibia ash increased (P〈0.10) and the Mn contents in above mentioned tissues decreased (P〈0.10) as dietary Fe level increased. The DMT1 mRNA levels of Fe-supplemented groups were lower (P〈0.10) than that of the control, and the FPNI mRNA level of Fe-supplemented 500 mg/kg group was lower (P〈0.10) than those of the control and the other two Fe-supplemented groups. The Mn absorption rates of Fe-supplemented groups were lower (P〈0.10) than that of the control. These results suggested that dietary Fe might decrease the absorption and deposition of Mn through the down-regulations of DMT1 and FPNI gene expression in duodenal mucosa of broilers.
