摘  要:目的:检测早期生长反应因子-1(Egr-1)mRNA在大鼠腹主动脉瘤(AAA)模型组织中的表达情况,以探讨其在腹主动脉瘤发病机制中的作用。方法:20只SD雄性大鼠,随机分成2组,一组为对照组,另一组为实验组,实验组为弹力蛋白酶灌注(20U/m L)1m L,持续30min。分别术前,术后14d测量腹主动脉直径。应用q-PCR方法检测动脉瘤中早期生长反应因子-1 mRNA的表达,数据均以x±s或者配对t检验进行两组之间的差异分析。结果:实验组动物模型均已达到腹主动脉瘤诊断标准(对照组:1.400±0.033,实照组:2.950±0.171,每组10只,P<0.05)。与对照组相比,实验组Egr-1mRNA的表达明显高于对照组(对照组:1.026±0.122,实验组:6.177±0.721,P<0.05)。结论:早期生长反应因子-1 mRNA在大鼠腹主动脉瘤中的表达明显高于对照组,说明早期生长反应因子-1可能在介导腹主动脉瘤的形成中发挥重要作用。Objective: To investigate the relationship between early growth response factor - 1 ( Egr - 1 ) and the formation of abdominal aortic aneurysm (AAA) in rats. Methods :20 SD male rats were randomly divided into two groups: the control group and the experimental group. The experimental group was injected with elastase (20U/mL) lml for 30min. Aortic diameter was measured before and after performance of elastase perfusion and before they were killed at 14th Day. The change of Egr - 1 mRNA was detected by q - PCR. The data were analyzed with x ± s or paired t test for the difference between the two groups. Results: The animal model of experimental group had accorded with the standard of diagnosis of abdominal aortic aneurysm ( control group : 1. 400 ±0. 033 ; the control group: 2.950 ±0. 171, each group of 10, P 〈 0. 05). Compared with the control group, the expression of Egr - 1 mRNA in the experimental group was significantly higher than that in the control group ( control group : 1. 026 ± 0. 122, the experimental group : 6. 177 ± 0.721, P 〈 0. 05 ). Conclusion: The changing of Egr - 1 mRNA expression is parallel to AAA pathogenesis. It suggested that Egr- 1 may affect on AAA formation.
