摘  要:目的探讨支气管哮喘患者血清中miRNAs表达谱改变情况,从中发现与支气管哮喘相关的miRNAs。方法选择2014年8月~2015年10月在解放军四二一医院确诊的48例哮喘急性发作期患者为哮喘组,另选择同期医院体检科健康体检者40例为对照组。通过miRNA表达谱芯片技术,对两组血清中差异表达的miRNAs进行检测,筛选得到的miRNAs通过实时荧光定量RT-PCR验证。结果哮喘组患者血清miR-155、miR-150和miR-145的表达明显高于对照组(P<0.05);而miR-34b-5p的表达则明显低于对照组(P<0.05)。结论支气管哮喘患者血清中miRNAs表达谱发生明显变化,提示血清miRNAs可能作为是支气管哮喘的检测指标,也可能作为未来哮喘治疗的新靶点。Objective To investigate changes of expression profile of miRNAs in serum of patients with bronchial asth- ma, in order to discover the miRNAs related to bronchial asthma. Methods From August 2014 to October 2015, in Hospital No.421 of PLA, 48 patients diagnosed with acute asthma were selected as asthma group, 40 healthy persons had physical examination at the same period were selected as control group. Expression of miRNAs in serum of patients with bronchial asthma and healthy controls were detedted by miRNA microarray technique, and miRNAs obtained by screening were validated by real time fluorescent quantitative RT-PCR. Results The expression of miR-155, miR-150 and miR-14 in serum of asthma group were higher than control group (P 〈 0.05); and the expression of miR-34b-Sp was lower than control group (P 〈 0.05). Conclusion The expressing profile of miRNAs in serum of patients with bronchial asthma changed significantly, indicating that serum miRNAs can be used as detecting parameter for bronchial asthma and may also serve as new targets in the treatment of asthma in the future.
