摘　　要：目的探讨华支睾吸虫颗粒蛋白(CsGRN)在虫体和宿主中的表达。方法使用荧光定量PCR(qRT-PCR)检测华支睾吸虫成虫、囊蚴和虫卵及猫的肝脏、胆管组织和全血CsGRN基因mRNA表达水平;免疫组化检测CsGRN在成虫及宿主猫、人和实验感染的Balb/c小鼠肝脏中的定位。结果 CsGRN基因mRNA在虫体的各阶段都有表达,在感染的猫肝和胆管组织中高表达;CsGRN主要定位在成虫的睾丸、外膜和虫卵,在宿主人、猫和小鼠中主要分布在胆管上皮和肝细胞。结论 CsGRN可能参与虫体的生长发育,它作为分泌排泄抗原之一可能损害肝胆管组织,有进一步研究价值。Objective To explore the expression of Clonorchis sinensis granulin（CsGRN） in the parasite and host.Methods The relative mRNA level of CsGRN in adult worms,metacercaria and eggs of Clonorchis sinensis（C. sinensis）as well as the hepatic tissues,bile duct tissues and whole blood of infected cats was detected by qRT-PCR. The distribution of CsGRN in liver of cancer patients,infected cats and experimental Balb/c mice was determined by immunohistochemistry.Results CsGRN was transcribed in all stages of C. sinensis and the expression levels of CsGRN were high in hepatic tissues and bile duct tissues of infected cats. CsGRN was mainly located in testis,tegument and eggs of adult worms as well as the biliary epithelium and hepatocytes of the host. Conclusion CsGRN may take part in the growth and development of C. sinensis. As an excretory-secretory antigen,CsGRN may be related to hepatic duct diseases,especially hepatocellular carcinoma and cholangiocarcinoma induced by C. sinensis.
