摘 要:目的探讨^(18)F-FDG PET/CT显像检测肺纤维化的诊断价值。方法选取12只雄性SD大鼠随机分为肺纤维化组和对照组,每组6只。肺纤维化组大鼠单次气管内注入博来霉素(5 mg/kg),对照组给予等量生理盐水。造模后28天两组大鼠进行肺组织^(18)F-FDG PET/CT显像,检测两组大鼠肺组织对^(18)F-FDG的摄取情况。显像后处死大鼠并取其左肺叶用于Masson染色,测定羟脯氨酸含量。结果肺纤维组肺组织摄取^(18)F-FDG的%ID/g值为(0.6±0.059),明显高于对照组的(0.095±0.005),差异有统计学意义(t=20.74,P<0.05)。肺纤维化组大鼠肺组织羟脯氨酸含量为(1.704±0.145)μg/mg,明显高于对照组的(1.066±0.057)μg/mg,差异有统计学意义(t=10.05,P<0.05)。Masson染色显示肺纤维化组大鼠肺组织纤维增生和胶原沉积,^(18)F-FDG PET/CT显像示肺纤维化组大鼠的肺纤维灶处可见明显放射性浓聚。肺纤维化组^(18)F-FDG摄取值与羟脯氨酸含量密切相关(R^2=0.9142,P<0.05)。结论^(18)F-FDG PET/CT显像可用于无创诊断肺纤维化。Objective To investigate the diagnostic value of^18F-FDG PET/CT imaging for the detection of pulmonary fibrosis.Methods 12 male SD rats were divided to two groups,including pulmonary fibrosis group and control group. The pulmonary fibrosis group received a single intratracheal instillation of saline containing 5 mg/kg bleomycin. Control group was treated with the same volume of saline. At day 28 after instillation,^18F-FDG PET/CT imaging was performed on two groups and measure the^18F-FDG uptake of lung tissues in two groups. After the PET/CT scans,all rats were sacrificed to harvest their left pulmonary lobe tissues for Masson staining,and the right pulmonary lobe tissues for measuring the hydroxyproline content. Results Compared with control group,pulmonary fibrosis group showed higher hydroxyproline content(P〈0.05).Pathological examination showed that obvious proliferation of fibroblasts and deposition of collagen occurred in lung tissues of pulmonary fibrosis group. The results indicated radioactivity concentration on fibrosis area of lung tissues in pulmonary fibrosis group.^18F-FDG uptake of lung tissues in the pulmonary fibrosis group was higher than that of control group(P〈0.05).Conclusion ^18F-FDG PET/CT imaging could be used for noninvasive diagnosis of pulmonary fibrosis.
