肾白宁对糖尿病肾病大鼠ACE2蛋白及基因表达的影响

摘　　要：目的:观察肾白宁对糖尿病肾病大鼠ACE2蛋白及基因表达的影响。方法:采用左侧肾脏摘除+STZ诱发DN大鼠模型,将成模大鼠随机分为4组:假手术组、模型组、对照组、中药组。中药组将肾白宁浸膏溶于生理盐水,配制成混悬液,予以2.97 g·(kg·d)^(-1),2 m L混悬液灌胃;对照组将盐酸贝那普利溶于生理盐水,配制成混悬液,予以中等剂量0.9mg·(kg·d)^(-1),2 m L混悬液灌胃;模型组及假手术组予以等体积生理盐水,1次·d-1,灌胃,观察12周。检测大鼠肾脏组织ACE2蛋白及基因表达。结果:与假手术组比较,模型组、对照组、中药组ACE2蛋白及基因表达水平明显降低,差异具有统计学意义(P<0.01)。与模型组比较,对照组、中药组ACE2蛋白及基因表达水平明显升高,差异有统计学意义(P<0.05或P<0.01)。与对照组比较,中药组ACE2蛋白表达水平明显升高,差异具有统计学意义(P<0.05);ACE2基因表达水平升高,但差异无统计学意义(P>0.05)。结论:肾白宁能明显升高糖尿病肾病模型大鼠肾脏组织ACE2蛋白及基因表达,从而起到拮抗肾脏纤维化,延缓大鼠糖尿病肾病进展的作用。Objective： To observe the effects of Shenbaining（ SBN） on the ACE2 proteinum expression and mRNA level on streptozotocin（ STZ） induced diabetic nephropathy（ DN） rats. Methods： DN rat modeling was established by removing the left kidney of rat and the STZ-inducement. Rats are randomly divided into 4 groups： sham operation group,model group,control group and TCM treatment group. The rats in the TCM group are given 2. 97 g·kg^-1·d^-1 SBN（ dissolved in saline solution and was prepared for gavage）. The positive control group was given 0. 9 mg·kg^-1·d^-1 Benazepril（ dissolved in saline solution and prepared for gavage）. The model group and the sham operation group was given the same volume of saline solution. After 12 weeks,ACE2 proteinum expression and mRNA level of all groups were measured. Results： ACE2 protein expression and mRNA level was significanty inmodel group,control group and TCM treatment group compared with that of the sham operation group respectively（ P〈0. 01）.Compared with that of the model group,the ACE2 protein expression of the control group and the TCM treatment group is significantly higher（ P〈0. 05,P〈0. 01）. The ACE2 protein expression of the TCM treatment group was significantly higher than that of the control group（ P〈0. 05）. The mRNA level of each group was higher after intervention but the difference was not significant.Conclusions： SBN can increase ACE2 expression in protein and mRNA level in nephridial tissue,antagonize renal fibrosis and delay the progress of diabetic nephropathy in rats.
