精子不同处理方式对绵羊胞质内单精子注射效率的影响

摘　　要：胞质内单精子注射前对精子进行预处理是模拟精子受精过程的变化增强精子解聚能力。常用的精子处理方法有聚乙烯吡咯烷酮、二硫苏糖醇、冻融处理等。研究表明ICSI卵的卵裂率低的主要原因是精子解聚能力差。因此研究精子的处理方法提高ICSI的注射效率是十分必要的。该实验探讨了精子不同预处理方法对ICSI胚胎发育的影响。利用超声波细胞粉碎仪对精子做不同时间(3 s、6 s)的处理,观察ICSI胚胎发育情况,选择最佳的处理时间,然后利用超声波分别处理鲜精和冻精,比较不同精液对ICSI胚胎发育的影响。将精子分别利用超声波和碱处理,对比两种不同预处理方法对ICSI卵卵裂率和桑囊胚率的影响,结果表明:(1)超声波处理绵羊精子最佳的处理时间是6 s;(2)鲜精和冻精经超声波处理后对ICSI卵的卵裂率(39.1%vs 37.4%,P>0.05)和桑囊率(12.3%vs 13.2%,P>0.05)影响不显著;(3)冻精经碱处理后其ICSI卵的卵裂率(46.6%vs37.4%,P<0.05)和桑囊胚率(20.4%vs 13.2%,P<0.05)均显著高于超声波处理冻精。The aim of sperm pretreatment before sperm injection is to simulate the changes in the sperm fertilization process and to enhance the depolymerization capacity of the sperm.There are three kinds of sperm treatments that are frequently used： polyvinyl pyrrolidone（ PVP）,dithiothreitol（ DTT）,freezing-thawing process.Studies have shown that low sperm depolymerization is mainly caused by low cleavage rate.Therefore,it is necessary to study the sperm treatment method to improve the injection efficiency of ICSI.The effects of different pretreatment methods on the embryo development of ICSI were discussed.By comparing the effects of different ultrasonic treatment times（ 3 s,6 s） and spermatozoa（ fresh and frozen semen） on the cleavage rate of ICSI and the blastocyst rate,it was shown that（ 1） the optimal sheep sperm processing time was ultrasound 6seconds;（ 2） ultrasonic treatment of fresh and frozen ICSI semen showed no significant effect on the oocyte cleavage rate（ 39.1% vs 37.4%） and the morula/blatocyst rate（ 12.3% vs 13.2%）;（ 3） treated with（ Na OH and the oocyte cleavage rate was 46. 6%,vs 37. 4% and the morula/blastocyst rate was 20. 4% vs 13. 2%;P〈0.05,significanlly higher than those treated with ultrasonic waves.
