摘 要:以奶牛乳腺上皮细胞为细胞模型,用MTT法检测细胞的存活和生长情况,用不同浓度的黄酮溶液(0、100、200、400、600、800mg/L)刺激奶牛乳腺上皮细胞12h,再用相同浓度的黄酮溶液(400mg/L)作用于奶牛乳腺上皮细胞不同时间(1、4、9、12、16、24h),分别提取细胞总RNA,用荧光定量PCR法测定GHR、β-CN、IL-6、IL-8、TNF-α和IFN-γ泌乳相关基因的表达,研究葎草提取物总黄酮对乳腺上皮泌乳及炎性相关因子表达的影响。结果表明,试验组与空白组相比,奶牛乳腺上皮细胞中IFN-γ、TNF-α、IL-6、IL-8的表达水平降低(P<0.05);生长激素受体(GHR)、β-酪蛋白(β-CN)的表达量高于对照组(P<0.05)。说明葎草黄酮能上调奶牛乳腺上皮细胞β-CN和GHR的表达量,以浓度为400mg/L培养4h^12h的效果最佳。The aim of this study was to explore the effects of Humulus scandens flavonoids on related gene expressions of mammary gland epithelial cells. The cow mammary gland epithelial cells were used as cell model, and the survival and growth of the cells were detected by MTT method, the cow mammary gland epithelial cells were stimulated with different concentration of flavonoid solution (0, 100, 200, 400, 600, 800 mg/L) for 12 h, in the same time, the cow mammary gland epithelial cells were stimulated with the same concentration of flavonoid solution (400 mg/L) at different time (0 ,1, 4, 9, 12, 16, 24 h), and to- tal RNA was extracted from cells respectively. The expressions of GHR, β-CN, IL-6, IL-8, TNF-α and IFN-γ those are lactation-related genes were measured by the fluorescent quantitative PCR. The effects of total flavonoids from Humulus scandens extract on milk fat metabolism were studied. The experimental group was compared with the blank control group, the expression levels of IFN-γ, TNF-α, IL-6 and IL-8 in the cow mammary gland epithelial cells were decreased (P〈0.05) ; The expressions of GHR and β-CN were higher than that of the control group (P〈0.05) ; The results showed that the Hurnulus flavonoids concentration of 400 mg/L cultivated for 12 h, had a positive effect on the expression of β-CN; Hurnulus flavonoids concentration of 400 mg/L cultivated for 4 h, the expression of GHR had a role in promoting. This indicated that it plays an important role in the growth and development of mammary gland cells.
