不同炮制品配伍的续断散促进骨生长的比较研究

摘　　要：目的比较研究不同炮制品配伍的续断散促进骨生长的活性作用。方法采用MC3T3-E1成骨细胞模型,按药典法和《全国中药炮制规范》炮制续断和牛膝各种炮制品;采用MTT法测定各不同炮制品配伍组成的续断散的成骨细胞增殖率;采用碱性磷酸酶(ALP)试剂盒测定各不同炮制品配伍组成的续断散的成骨细胞ALP活力。结果与空白组相比,终浓度为1mg生药/m L时,除酒续断-生牛膝、酒续断-酒牛膝外,其余各种炮制品配伍的续断散在促进成骨细胞增殖方面分别具有显著性和极显著性差异(P<0.05,P<0.01),终浓度为0.25mg^1mg生药/m L的盐续断-酒牛膝在增强成骨细胞ALP活力方面分别具有显著性和极显著性差异(P<0.05,P<0.01)。结论不同炮制品配伍的续断散具有不同程度地促进骨生长的活性作用,且盐续断与酒牛膝为续断散的最佳配伍。OBJECTIVE To study the active effects of Xuduan Powder with different processed drugs on promo- ting bone growth. METHODS MC3T3-E1 osteoblasts were used as the model;according to Chinese pharmacopoeia and 〈 National standards for processing traditional Chinese medicine 〉 to processing Radix Dipsaci and Radix Achy- ranthis Bidentatae;the MC3T3-E1 osteoblast proliferation of each group was detected by MTr method;ALP activity of osteoblasts was determined by alkaline phosphatase assay. RESULTS Compared with the blank group, the final concentration of I mg crude dr μg·mL^-1, except the wine-processed Radix Dipsaci-crude Radix Achyranthis Bidenta- tae,the wine-processed Radix Dipsaci-the wine-processed Radix Achyranthis Bidentatae, the rest of the various Xudu- an Powder with different processed drugs in promoting osteoblast proliferation has a significant and very significant difference（P 〈0. 05 ,P 〈0. 01 ） ,the final concentration of 0. 25 - 1mg crude drμg·mL^-1,the salt-processed Radix Dipsaci-the wine-processed Radix Achyranthis Bidentatae in enhancing ALP activity of osteoblast has a significantly and very significant difference （ P 〈 0. 05, P 〈 0. 01 ）. CONCLUSION The results indicate that Xuduan Powder with different processed drugs have the effect of promoting bone growth in different degrees, and the salt-processed Radix Dipsaci-the wine-processed Radix Achyranthis Bidentatae is the best compatibility.
