摘 要:目的:探讨胃癌中医病理因素特征与胃癌组织抑癌基因ING1甲基化及CD133、CD44基因表达间的关系。方法:采集100例胃癌术前临床资料,进行中医病理因素的提取,探求其病理因素特征,并采用甲基化特异性PCR(MS-PCR)测其手术切除的胃癌组织和相应的癌旁组织中ING1甲基化水平,RT-PCR和免疫组织化学法测CD133、CD44水平,探讨ING1甲基化对CD133、CD44表达水平的影响,并采用二元logistic回归分析胃癌病理因素与ING1甲基化状况、CD133和CD44基因表达水平的相关性。结果:胃癌病理因素主要为气虚、痰、瘀、气滞;胃癌组织和癌旁组织ING1甲基化和CD133、CD44表达差异有显著性意义(P<0.05);胃癌组织ING1甲基化状况对CD133、CD44表达产生显著影响(P<0.05);ING1甲基化状况、CD133和CD44表达水平对胃癌中医病理因素气虚、痰、瘀产生显著影响(P<0.05)。结论:胃癌病理因素气虚、痰、瘀与ING1甲基化状况、CD133和CD44表达水平具有显著相关性。Objective : To explore the relationship between the characteristics of TCM pathological factors of gastric cancer and rumor suppressor gene ING1 methylation, as well as the expression of CD133 and CD44 in gastric cancer tissue. Methods : The preoperative clinical data of 100 cases of patients witb gastric cancer were collected, and the TCM pathological factors were extracted to explore their characteristics. The frequency of aberrant methylation of ING1 gene deteted by Methylation-specific PCR ( MSP )and expression levels of CD133 and CD44 gene measured respectively by RT-PCR and immunohistochemical method were to explore the effect of ING1 methylation on the expression of CD133 and CD44 gene in gastric tissue and matched adjacent tissue, and then the binary logistic regression model was used to analyzed the relationship between the characteristics of TCM pathological factors of gastric cancer and Iumor suppressor gene ING1 methylation, as well as the expression of CD133 and CD44. Results : The characteristics of TCM pathological factors of gastric cancer were Qi deficiency, phlegm, blood stasis and Qi stagnation; ING1 methylation and expression of CD133 and CD44 were significantly different between in gastric cancer tissues and adjacent tissues ( P〈0.05 ). The expression of CD133 and CD44 in gastric cancer tissue was significantly affected by ING1 methylation status ( P〈0.05 ). ING1 methylation status, expression levels of CD133 and CD44 in gastric cancer tissue had a significant impact on Qi deficiency, phlegm, stasis among TCM pathological factors of gastric cancer ( P〈0.05 ). Conclusion : The TCM pathological factors of gastric cancer are significantly correlated with ING1 methylation status, the expression of CD133 and CD44 gene.
