苏木乙酸乙酯提取物对心脏移植模型大鼠ICAM-1 mRNA表达影响

摘　　要：目的:通过观察苏木乙酸乙酯提取物对同种异位心脏移植急性排斥反应模型大鼠移植心肌ICAM-1 mRNA表达的影响,探讨其免疫抑制作用的机理,为苏木的新药研发提供重要的理论和实验依据。方法:以Wistar大鼠为供体,SD大鼠为受体,制备大鼠同种异位心脏移植模型,造模成功后,采用Real Time-PCR法测定ICAM-1 mRNA的表达。结果:与模型对照组相比,苏木组、Cs A组及苏木加半量Cs A组均能明显抑制ICAM-1 mRNA的表达,而苏木乙酸乙酯提取物及苏木乙酸乙酯提取物加半量Cs A抑制ICAM-1 mRNA表达的作用相当,且在抗排斥反应时,苏木乙酸乙酯提取物可以减少Cs A的用量。结论:在大鼠心脏移植模型中,苏木乙酸乙酯提取物能抑制同种异位心脏移植模型大鼠心肌ICAM-1 mRNA的表达,减轻移植心脏的急性排斥反应。Objective：Acute heart transplant rejection was investigated with rat model of heterotopic cardiac transplantation. Effect of ethyl acetate extracted from Caesalpinia sappan L. on tissue ICAM-1 was observed. The immune suppressive effect mechanism was investigated. These experiments were important theoretical and experimental bases for development of new drug with Caesalpinia sappan L.. Methods：With Wistar rats as donors and SD rats as recipients,expression of ICAM-1 mRNA was detected with Real TimePCR method. Results：After compared with the model control group,the expression of ICAM-1 mRNA was obviously suppressed in the treatment groups（including the Cs A group,Caesalpinia sappan L. added with half Cs A group and Caesalpinia sappan L. group）. The effect of the Caesalpinia sappan L. group on suppressing the expression of ICAM-1 mRNA was weaker than the Cs A group. The effect of the Cs A group on suppressing the expression of ICAM-1 mRNA was almost same with the Caesalpinia sappan L. added with half Cs A group. And in the anti rejection,Caesalpinia sappan L. ethyl acetate extract could reduce the dosage of Cs A. Conclusion：Ethyl acetate extracted from Caesalpinia sappan L. can suppress the expression of ICAM-1 mRNA of the cardiac muscle of the rat in the heterotopic cardiac transplantation,reduce acute rejection of transplanted heart.
