摘 要:目的研制一种新型的具有分子靶向显像潜能的高掺钆量的介孔二氧化硅纳米粒子用于MRI。方法采用反向微乳液法将钆喷酸掺杂到介孔二氧化硅(MSNs)孔道中,制备钆喷酸掺杂的介孔二氧化硅纳米粒子(GdMSNs)。ICP-AES检测纳米粒的含钆量;透射电镜观察纳米粒的形貌特征;3.0 T MRI系统体外观察纳米粒。结果成功制备出Gd-MSNs,电感耦合等离子体发射光谱仪测得掺钆量百分比达12.11%;制备的Gd-MSNs形态呈圆球形,直径约50~70 nm。MRI显示Gd-MSNs能显著缩短T_1和T_2的弛豫时间,表现为短T_1、短T_2的信号特点。结论成功制备的单分散、易修饰、高载药量的新型掺钆纳米材料,有望成为一种新型的多功能靶向MRI纳米对比剂。Objective To synthesize a high gadolinium-doped mesoporous silica nanospheres(Gd-MSNs) with a potential molecular target imaging as a novel MRI contrast agent.Methods Reverse microemulsion method was used to dope gadopentetic acid into the channels of MSNs to synthesize Gd-MSNs.Gadolinium content was detected by ICP-AES.The morphology and size of nanoparticles was detected by transmission electron microscope and nanoparticle dilutions was scanned by 3.0T MRI system.Results Gd-MSNs with high Gadolinium content percentage of 12.11% was synthesized.The globular-shaped Gd-MSNs was approximately 50 nm to 70 nm in diameter.MRI showed that Gd-MSNs can significantly shorten the T1 and T2relaxation time.Gd-MSNs showed short T1 and short T2 signal features.Conclusion The successful preparation of the gadolinium-doped nanoparticles,which was monodisperse,easy to modify and has high Gadolinium loadings,is expected to become a novel multifunctional targeted highly efficient MRI nano contrast agent.
