摘  要:[目的]在云南省昆明市植物园中发现症状表现为叶片褪绿和环斑的天竺葵,怀疑其为番茄斑萎病毒属病毒侵染。[方法]将病叶汁液摩擦接种至本氏烟,显症后取样与原样一起用3种番茄斑萎病毒属病毒的抗体采用Dot-blot ELISA方法进行检测,并用RT-PCR方法对本氏烟叶片进行检测。[结果]接种本氏烟的系统叶片在7 d后表现典型的花叶、卷曲和枯斑等症状,天竺葵病叶和接种显症的本氏烟系统叶Dot-blot ELISA和本氏烟叶片RT-PCR检测结果均表明其病原为番茄斑萎病毒(Tomato spotted wilt virus,TSWV)。RT-PCR产物的测序结果显示其N基因与TSWV-LE分离物核苷酸相似性达99.74%。[结论]天竺葵病样为TSWV侵染所致,这是首次在中国天竺葵上发现TSWV,对该病毒的防治工作具有参考价值。[Objectives] Some geraniums (Pelargonium hortorum)showing chlorotic and ring spots, which look like being infected by tospoviruses ,were found in Kunming Botanical Garden, Yunnan Province. [ Methods ] The crude extracts of sampled geranium leaves were rub-inoculated to Nicotiana benthamiana plants and the systemic leaves of the inoculated plants were detected together with the original sample by Dot-blot ELISA after symptom observation. The symptomatic N. benthamiana leaves were further detected by RT-PCR. [ Results ] The systemic leaves of inoculated N. benthamiana plants showed obvious mosaic, leaf curling and necrosis after seven days post inoculation. The results of Dot-blot ELISA and RT-PCR detection indicated that they were infected by Tomato spotted wilt virus (TSWV). The RT-PCR products amplified by specific primers of TSWV N gene were sequenced and the sequence analysis revealed that it shared 99.74% identity with TSWV-LE isolate at the nucleotide level. [ Conclusions] The data above suggests that the sampled geraniums showing ring spots are infected by TSWV,which is the first report in China on the infection of TSWV in geraniums and useful for the virus control.
