摘  要:目的建立微波消解-石墨炉原子吸收光谱法测定血和尿中铅和镉含量的方法,为临床铅镉中毒提供诊疗依据。方法取0.50 ml血液和尿液,在HNO3/H2O2消解剂中微波消解5 min,然后以硫酸铵-磷酸二氢铵混合溶液为基体改进剂,采用石墨炉原子吸收光谱法对消解液中的铅镉含量进行测定。结果铅、镉在分别在0.0~160μg/L和0.0~16.0μg/L范围内线性良好(相关系数分别为铅0.9993,镉0.9987),方法检出限(3 s/b)分别为0.36μg/L和3.24μg/L。加标回收率在97.8%~103.0%之间,测定值的相对标准偏差(n=6)在1.76%~6.81%之间。结论该方法快速灵敏,精密度和准确度均符合要求,可为临床诊断铅镉中毒提供一个简便准确的检测方法。Objective The study aimed to establish a method for determination oflead and cadmium contents in blood and urineby microwave digestion-graphite furnace atomic absorption spectrometry, so as to provide a reference for clinical diagnosis and treatment of lead and cadmium poisoning. Methods 0.50mL of blood and urine samples were subjected to microwave digestion for 5min in HNO3/H2O2 digestion solution. Ammonium sulfate and ammonium dihydrogen phosphate mixture was used as matrix modifier for graphite furnace atomic absorption spectrometry to measure the lead and cadmium contents in the digestion solution. Results The lead and cadmium contents showed good linearity within the ranges of 0.0- 160μg/L and 0.0-16.0μg/L, respectively (the correlation coefficient was 0.9993 for lead and 0.9987 for cadmium). The detection limits (3s/b)of the method were0.36μg/L and 3.24μg/L, respectively. Average recovery rate of thesamples ranged between 97.8%and 103.0%,and the relative standard deviation (RSD) (n=6) was 1.76%-6.81%. Conclusion The method isfastand meets the requirements for precision and accuracy, providing a simple and accurate determination method for clinical diagnosis of lead and cadmium poisoning.
