摘 要:目的研究海蛇风湿胶囊抗风湿的药效作用。方法取SD大鼠60只,随机分为6组,每组10只,即空白对照组,模型组,海蛇风湿胶囊低、中、高剂量组(82.3,164.5,329.0 mg·kg^(-1)),雷公藤多苷片组(7.9 mg·kg^(-1)),分组给药,第5次给药后制备大鼠佐剂性关节炎模型,观察大鼠足趾肿胀度和炎症抑制率;取小鼠50只,随机分为5组,正常对照组,海蛇风湿胶囊低、中、高剂量组(156.6,313.2,626.4 mg·kg^(-1)),阳性药物(阿司匹林/吲哚美辛)组(17.1 mg·kg^(-1)),小鼠背部皮下注射2%灭菌琼脂,制备小鼠琼脂致肉芽肿实验,右后脚趾部皮下注射2.5%的福尔马林液制备镇痛实验,比较各组肉芽肿质量和疼痛评分。腹腔注射0.7%的冰醋酸制备小鼠扭体实验,观察发生扭体反应的次数和潜伏期;小鼠热水甩尾法镇痛实验,测定给药前后的痛阈值。结果海蛇风湿胶囊对大鼠佐剂性关节炎均具有预防作用,致炎后6 h,海蛇风湿胶囊高剂量组的炎症抑制率达到52.73%;能抑制小鼠琼脂肉芽肿的形成并在福尔马林镇痛实验中,表现出良好的镇痛作用,高剂量组的疼痛总评分均值为100.60;明显减少小鼠扭体反应次数(P<0.01);在热水甩尾实验中明显提高小鼠痛阈值(P<0.01)。结论海蛇风湿胶囊具有良好的抗风湿作用,且以高剂量组为最佳。Objective To research the anti-rheumatism effect of Haishe Fengshi Capsules (HFC). Methods Sixty Sl) rats were randomly divided into 6 groups, namely blank control group, model group, low-, medium- and high-dose HFC groups(82.3, 164.5, 329.0 mg·kg^-1 respectively) and triptolide group(7.9mg·kg^-1), 10 rats in each group. After administration with the corresponding medicine for 5 d, adjuvant arthritis(AA) rat model was established for the observation of pedal swelling and inflammation-inhibition rate. In another experiment, 50 mice were randomly divided into 5 groups, namely normal control group, low-, medium- and high-dose groups (156.6, 313.2, 626.4 mg·kg^-1 respectively) and positive drug group (aspirin/indomethacin 17.1 mg·kg^-1). The mouse agar-induced granuloma model was prepared by subcutaneous injection of 2 % sterilized agar. Analgesic experiment was carried out by subcutaneous injection of 2.5 % of the formalin solution into the right posterior toes, and then the granuloma mass and pain scores were compared. Mouse writhing model induced by intraperitoneal injection of 0.7 % glacial acetic acid was used to observe the number of writhing reaction and latency. Hot water-induced pain test was used to measure the pain threshold of mice before and after administration. The above experiments were performed for studying the anti-rheumatic effect of HFC. Results HFC had preventive effect on AA rats. After inflammation for 6 hours, the inflammation-inhibition rate in high-dose HFC group was 52.73 %. HFC inhibited the formation of agar-induced granuloma in mice and showed good analgesic effect in formalin-induced pain test, with the average pain scores of 100.60 in high-dose HFC group. HFC significantly reduced the number of writhing reaction in mice(P 〈 0.01) and increased the mouse pain threshold in hot water-induced pain test (P 〈 0.01 ). Conclusion HFC have certain anti-inflammatory and analgesic effect, and high-dose HFC have the strongest effect.
