摘  要:目的:进行体外透皮吸收试验,考察磷脂固体分散技术对盐酸小檗碱体外透皮能力的影响。方法:以离体小鼠腹部皮肤为透皮屏障,采用改良Franz扩散池装置进行透皮吸收实验,采用紫外分光光度法测定透皮接收液中小檗碱含量,比较盐酸小檗碱及其磷脂固体分散体透皮能力的差异,评价磷脂固体分散技术对小檗碱透皮吸收的促进作用。结果:高、中、低浓度盐酸小檗碱磷脂固体分散体的累计透皮量均远高于同等浓度盐酸小檗碱。结论:磷脂固体分散技术可提高盐酸小檗碱的透皮能力,盐酸小檗碱磷脂固体分散体可用于透皮吸收制剂的研发。OBJECTIVE To study the percutaneous penetration ability of berberine hydrochloride with phospholipid solid dispersion method in a transdermal absorption experiment in vitro.METHODS Ultraviolet spectrophotometry was established to determine berberine content in transdermal receiving liquid with abdominal skin of mouse as the transdermal barrier,adopting modified Franz instument was used to perform transdermal absorption test,to compare the percutaneous penetration ability of berberine hydrochloride and its phospholipid solid dispersions,and evaluate the promoting effects of phospholipid solid dispersion on transdermal absorption of berberine.RESULTS High,medium and low concentrations of berberine hydrochloride phospholipid solid dispersions had obviously higher cumulative transdermal rates than that of berberine hydrochloride at the same concentration.CONCLUSION Phospholipid solid dispersion method can significantly increase the transdermal absorption of berberine hydrochloride,and berberine hydrochloride phospholipid solid dispersion preparation can be used in research and development of the transdermal absorption preparation.
