槲寄生滴眼液治疗兔围绝经期干眼症的应用研究

摘　　要：目的:探究槲寄生滴眼液对治疗围绝经期雌兔干眼症的效果。方法:取健康雌兔36只将24只雌兔行双侧卵巢切除术,处理后,将这24只受试兔按照随机数字表法分为2组(每组12只,均为右眼):对生理盐水组(A组)、槲寄生滴眼液组(B组),连续滴眼2个月,每日4次。剩余的12只雌兔(C组)打开腹腔,不切除卵巢,眼部不进行任何处理,作为假手术组。分别于手术前、手术后1周、2周、1个月及2个月对受试兔行基础泪液分泌(SIT)检查、角膜荧光素染色(FL)评分、泪液蛋白测定及角膜共聚焦显微镜扫描。结果:A、B2组干预前SIT、FL相比,差异无统计学意义(P>0.05),与假手术组比较,差异均有统计学意义(均P<0.05);干预2个月后,假手术组SIT、FL评分较干预前没有明显变化(P>0.05),而A、B组SIT、FL评分较干预前变化明显,差异均有统计学意义(P<0.05)。干预2个月后,B组与假手术组相比,差异无统计学意义(P>0.05),而A组与假手术组相比,差异有统计学意义(P<0.05)。A、B2组各时间点SIT、FL评分相比,差异均有统计学意义(P<0.05)。A、B2组干预2个月后泪液总蛋白量、乳铁蛋白、溶菌酶及淀粉酶活性相比,差异均有统计学意义(P<0.05)。经角膜共焦显微镜显示:B组角膜上皮下神经及角膜基质层神经干较直,与正常雌兔上皮下神经比较数量稍有减少。而A组角膜上皮下神经及角膜基质层神经干弯曲,数量明显减少。结论:槲寄生滴眼液可有效减轻围绝经期雌兔干眼症的症状和体征,应用于围绝经期女性干眼症治疗有一定的临床意义。OBJECTIVE To investigate effects of mistletoe eye drops in treating perimenopausal xerophthalmia in female rabbits.METHODS Thirty-six healthy female rabbits were chosen,24 female rabbits were cut off bilateral ovaries,then the perimenopausal dry eye models were established and randomly divided into 2 groups：the normal saline group（group A）,the mistletoe eye drops group（group B）,sham operation group（12 eyes of the other 12 rabbits,group C）.For animals in group C,abdominal cavity was opened but ovaries were not removed,no processing procedure was performed and group C was used as a control group.Schirmer＇s test（SIT）,corneal fluorescein（FL）,tear proteins,and corneal confocal microscope were performed before intervention and 1 week,2 weeks,1month,2 months after intervention.RESULTS There was no significant difference in SIT and FL between group A and B（P〉0.05）.After 2 months of intervention,SIT and FL in the sham operation group did not change（P〉0.05）.Compared to those before intervention,SIT and FL slightly changed in group B,but with statistical significance（P〈0.05）,and more significantly changed in group A（P〈0.05）.Compared with the sham operation group,SIT and FL of group A were significantly different（P〈0.05）.There were statistically differences in SIT and FL at each time point between group A and B（P〈0.05）.After 2 months of intervention,tear protein,lactoferrin,lysozyme and amylase activity had statistical difference compared in group A and B（P〈0.05）.The corneal confocal microscopy showed nerve was straight under corneal epithelium and within the matrix in group B,compared with normal female rabbits,the number of nerve slightly reduced.While nerve was straight under corneal epithelium and within the matrix in group A,the number of nerve significantly decreased.CONCLUSION Mistletoe eye drops can effectively alleviate the signs and symptoms of dry eyes in perimenopausal female rabbits,which provides a theoretical bassi to treat dry eyes in perimenop
