摘 要:目的:探讨表达IL-29的重组新城疫病毒(recombinant Newcastle disease virus IL-29,r NDV IL-29)对肺腺癌A549细胞增殖、迁移和细胞凋亡的影响。方法:实验分为r NDV IL-29组(r NDV IL-29感染A549细胞)、NDV组(NDV感染A549细胞)和对照组(PBS)。用Western blotting、RT-PCR、免疫荧光法检测IL-29蛋白及其mRNA的表达;MTT、克隆形成实验及划痕实验检测r NDV IL-29对A549细胞增殖、迁移能力的影响;流式细胞术及Western blotting检测r NDV IL-29对A549细胞凋亡的影响。结果:与NDV组和对照组相比,r NDV IL-29组细胞:(1)IL-29蛋白及其mRNA表达水平显著升高;(2)细胞增殖抑制率明显升高[(56.48±11.89)%vs(42.18±6.18)%,P<0.05],细胞迁移率明显下降[(5.00±4.00)%vs(32.43±3.71)%、(84.57±3.96)%,均P<0.05];(3)Caspase3表达水平明显上调[(29.67±1.53)%vs(18.33±3.06)%、(7.00±6.08)%,均P<0.05],Bcl-2/bax表达水平明显下调[(12.00±2.65)%vs(42.33±5.86)%、(67.00±6.25)%,均P<0.05],细胞凋亡增多。结论:r NDV IL-29抑制肺腺癌A549细胞增殖、迁移,并且促进其凋亡,提示r NDV IL-29可能成为一种很有潜力的抗癌药物。Objective: To investigate the effects of recombinant Newcastle disease virus (rNDV) expressing IL-29(rNDV IL-29)on the proliferation, migration and apoptosis of lung adenocarcinoma A549 cells. Methods: A549 cells were treated with rNDV IL-29, NDV and PBS respectively, namely rNDV IL-29 group, NDV group and control group. The protein and gene expression of IL-29 in A549 cells were examined by RT-PCR, immunofluorescence and Western blotting; MTT assay, colony formation assay and scratch assay were used to evaluate the effect of rNDV IL-29 on proliferation and migration ability of A549 cells; Flow cytometry and Western blotting were used to examine the effect of rNDV IL-29 on apoptosis of A549 cells. Results:Compared with NDV group and control group, (1) IL-29 protein and gene expressions were dramatically increased in rNDV IL-29 group. (2) the rate of cell proliferation in rNDV IL-29 group was significantly inhibited ([56.48±11.89]% vs [42.18±6.18]%, P〈0.05), and migration ability of A549 cells in rNDV IL-29 group was statistically decreased ([5.00±4.00]% vs [32.43±3.71]%, [84.57±3.96]%, all P〈0.05). (3) the expression of caspase3 in rNDV IL-29 group was significantly up-regulated ([29.67±1.53]% vs [18.33±3.06]%, [7.0±6.08]%, all P〈0.05), while Bcl-2/bax level was remarkably reduced ([12.00±2.65]% vs [42.33±586]%, [67.00±6.25]%, all P〈0.05]. Conclusion: rNDV IL-29 inhibits the proliferation and migration of A549 cells and promotes apoptosis of A549, suggesting that rNDV IL-29 may be served as a potential anticancer drug.
