摘  要:目的:建立高效液相色谱法同时测定不同产地掌叶大黄中10个蒽醌类成分(芦荟大黄素-8-O-葡萄糖苷、大黄酸-8-O-β-D-葡萄糖苷、大黄酚-8-O-β-D-葡萄糖苷、大黄素-8-O-葡萄糖苷、大黄素甲醚-8-O-β-D-葡萄糖苷和芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚)。方法:采用C_(18)色谱柱(250 mm×4.6 mm,5μm),甲醇-0.1%磷酸水为流动相,梯度洗脱,流速为1.0 mL·min^(-1),检测波长254 nm,柱温25℃。结果:10个成分在测定范围内线性关系较好(r>0.999),芦荟大黄素-8-O-葡萄糖苷、大黄酸-8-O-β-D-葡萄糖苷、大黄酚-8-O-β-D-葡萄糖苷、大黄素-8-O-葡萄糖苷、大黄素甲醚-8-O-β-D-葡萄糖苷和芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚平均回收率(n=6)分别为99.0%、99.2%、99.0%、99.2%、99.0%、99.2%、98.8%、99.4%、98.7%、99.6%;不同产地大黄药材中上述10个蒽醌类成分含量差异明显,含量范围分别为0.044%~0.130%、0.348%~1.686%、0.028%~0.183%、0.164%~0.352%、0~0.116%、0.095%~0.161%、0.294%~0.411%、0.157%~0.265%、0.275%~0.321%、0.064%~0.150%。结论:本文建立的方法样品处理简便,分析灵敏,能为大黄药材的质量评价提供参考。Objective: To establish simultaneous determination method of ten anthraquinones ( aloe-emodin-8- O-glucopyranoside, rhein-8-O-glucopyranoside, chryohol-8-O- β -D-glucoyroide, emodin-8-O-glueoside,physcion-8-O- β -D-monoglucoside, aloe-emodin, rhein, emodin, chrysophanol and physcion ) in Rheum palmatum L. from different habitants by HPLC. Methods: Cls column ( 250 mm×4.6 mm, 5 μm )was adopted, the mobile phase was a mixture of methanol and 0.1% phosphoric acid at the flow rate of 1.0 mL· min-1, the determination wavelength was set at 254 rim, the column temperature was 25 ℃ and the injection volume was 10 μL. Results: In the determination range, the linear relationship of ten components was good, and the recoveries of aloe-emodin-8-O-glucopyranoside, rhein-8-O-glucopyranoside, chryohol-8-O- β-D-glucoyroide, emodin-8- O-glucoside, physcion-8-O- β -D-monoglucoside, aloe-emodin, rhein, emodin, chrysophanol and physcion were 99.0%, 99.2%, 99.0%, 99.2%, 99.0%, 99.2%, 98.8%, 99.4%, 98.7% and 99.6%, respectively. The contents of ten anthraquinones in Rhei Radix et Rhizoma from different habitants were 0.044%-0.130%, 0.348%-1.686%, 0.028%-0.183%, 0.164%-0.352%, 0-0.116%, 0.095%-0.161%, 0.294%-0.411%, 0.157%-0.265%, 0.275%- 0.321% and 0.064%-0.150%, which showed remarkable difference. Conclusion: The proposed method was simple and sensitive, thus providing reference for the quality evaluation of Rheum palmatum L.
