摘  要:目的:建立同时测定葫芦素B和E含量的测定方法,考察^(60)Co射线辐照灭菌对5种葫芦科植物中葫芦素B和E含量的影响。方法:采用TC-C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)和0.1%醋酸溶液(B)系统,梯度洗脱,流速为1.0 mL·min^(-1),检测波长234 nm,柱温30℃;分别选择辐照剂量为2、5、10 kGy对葫芦科植物进行辐照,比较辐照前后活性成分含量的变化,并进行配对t检验考察其显著情况。对甜瓜蒂、南瓜蒂、栝楼、雪胆和木鳖子辐照前后的样品进行葫芦素B和E测定。结果:葫芦素B和E分别在0.548~8.765μg和0.407~6.504μg范围内有良好线性,平均加样回收率分别为99.2%~99.8%和99.3%~99.6%;葫芦素B含量辐照前后无显著性差异(p>0.05),而辐照剂量超过5 kGy时,葫芦素E辐照前后含量变化显著(p<0.05)。经5、10 kGy辐照后,甜瓜蒂、南瓜蒂、栝楼、雪胆和木鳖子中葫芦素B含量分别降低14.67%、65.08%、35.16%、30.40%和43.52%。结论:为不影响活性成分含量,采用60Co射线对葫芦科植物进行灭菌时,辐照剂量不宜超过5 kGy。Objective: To establish an HPLC method for simultaneous determination of cucurbitacin B and E, and to investigate the influence of 60Co- γ ray irradiation on the contents of eucurbitacin B and E in five cucurbitaceous plants. Methods: Separation was performed on a TC-Cls column ( 250 mm× 4.6 mm, 5 μm ). The mobile phase consisted of acetonitrile ( A ) and 0.1% acetic acid ( B ) with a gradient elution at the flow rate of 1.0 mL· min-1. The detection wavelength was 234 nm and column temperature was 30 , The Cucurbitaceae plants were irradiated at the dose of 2, 5 and 10 kGy, respectively. Then contents of the active ingredients before and after irradiation were compared by t-test. Cucurbitaein B and E in Pedicellus melo, Cucurbita moschata, Trichosanthes kirilowii, Hemsleya amabilis and Momordica cochinchinensis before and after irradiation were determined. Results: The linear range of cueurbitaein B and cucurbitacin E were 0.548-8.765 μg and 0.407-6.504 μg, and the average recoveries were 99.2-99.8% and 99.3-99.6%. There was no significant difference (p〈0.05)between contents of cucurbitacin E in samples before and after irradiation. But significant difference was found between contents of cucurbitaein B in samples irradiated at dose exceeding 5 kGy. The contents of Cucurbitacin B in Pedicellus melo, Cucurbita moschata, Trichosanthes kirilowii, Hemsleya amabilis and Momordica cochinchirtensis were decreased by 14.67%, 65.08%, 35.16%, 30.40% and 43.52% respectively after irradiation at dose of 5 kGy and 10 kGy. Conclusion: To avoid the variation of the active ingredients, cucurbitaceous plants shouldn't be irradiated at dose exceeding 5 kGy.
