赖氨酸修饰硅胶颗粒担载有机聚合物整体柱的制备及其在液相色谱中的应用

摘  要:在预先填充了5μm硅胶颗粒的150μm内径的石英毛细管内,采用原位聚合甲基丙烯酸缩水甘油酯和季戊四醇三丙烯酸酯的方法制备了多孔整体连续床,再用L-赖氨酸衍生化,得到一种可带两性电荷、具备多种分离机理的有机与无机混合型毛细管液相色谱整体柱.依据扫描电镜图和压力对流速关系曲线确定了整体柱的最佳制备条件,考察了整体柱的渗透性、机械稳定性、溶胀行为和亲水性,并利用离子交换机理分离了一组无机阴离子,利用亲水作用和离子交换混合机理分离了一组核苷酸化合物.A novel silica--particle--supported zwitterionic polymer monolith for capillary liquid chroma- tography was prepared by in situ coating 5 μm bare silica particles with the copolymers of glycidyl methac- rylate and pentaerythritol triacrylate, and further modifying with L--lysine inside a 150 μm i. d. fused cap- illary. The optimal conditions for preparation of the column were established based on SEM images and pressure--vs-flowrate curves. The prepared column was evaluated in terms of morphology, permeabili- ty, mechanical stability, swelling--shrinking behavior and hydrophilicity. Finally, a short section of the column was successfully applied to the separation of several inorganic anions based on ion exchange interac- tion and to a mixture consisting of four nucleotides based on a mixed mode of ion exchange and hydrophilic interaction.
