摘  要:目的:构建缺乏免疫反应的新型主动脉夹层(AD)动物模型,并研究不同的造模药物剂量,为探讨人源性活性物质在AD发生发展中的作用奠定实验基础。方法:将52只裸鼠随机分为实验组(42只)和对照组(10只)。实验组42只随机分为A、B、C、D四组,使用不同剂量给予β-氨基丙腈(BAPN)饮水28d和血管紧张素Ⅱ(Ang Ⅱ)皮下灌泵24h;对照组正常饮水28d。每7天测量一次体质量,观察体质量改变;第28天处死存活裸鼠,观察血管直观变化,使用HE和EVG染色观察形态学改变,并计算各组AD形成率及破裂率。结果:对照组无AD形成,实验组部分小鼠可见明显的主动脉扩张及壁内血肿,血管可见内膜撕裂、血细胞进入管壁,弹力纤维紊乱或断裂,证实形成AD。实验A、B、C、D组形成率分别为50%、90%、70%及80%,破裂率分别为41.67%、80%、20%及70%。结论:成功构建简单、方便的主动脉夹层免疫缺陷动物模型,并形成不同的形成、破裂率,以适应不同研究方案。Objective: To establish a lack of immune response to the new aortic dissection (AD) ani- mal model and to study the different dose of modeling drugs. Try to explor the role of human active substances in the development of AD. Methods: Fifty-two nude mice were randomly divided into two groups : experimental group (n = 42) and control group (n = 10). Experimental group: different dose of BAPN in drinking water for 28 ways and infused with different dose of AngII subcutaneously using a micro-osmotic pump for 24h. Control group:normal drinking water for 28 days. Observing the changes of body weight by measured every 7 days and blood vessels by sacrificed at 28 days. HE and EVG staining were used to observe the morphological changes, AD formation rate and rupture rate were analyzed. Results: No AD formation was observed in the control group. Some of the mice in the experimental group showed obvious dilatation of the aorta and intramural hemato- ma, blood vessels showed intimal tear, blood cells into the wall and elastic fiber disorder or fracture, which confirmed the formation of AD. The AD formation rates of Experimental group A, B, C, D were 50% , 90% , 70%, 80% and the rupture rates were 41.67%, 80%, 20%, 70% respectively. Conclusion: We have suc- cessfully established the models of aortic dissection in the immunodeficient animal. Different formation and rup- ture rate to adapt to different research programs.
