摘 要:目的探讨不同药物浓度的蜜炙紫菀水煎剂在体外实验中对乳腺癌MCF-7细胞的增殖、周期、迁移、凋亡的影响。方法通过MTT实验观察药物的处理对乳腺癌MCF-7细胞的生长与增殖情况的影响,采用流式细胞术研究蜜炙紫菀水煎剂对乳腺癌MCF-7细胞的增殖、周期和凋亡的影响,通过划痕实验观察蜜炙紫菀水煎剂对乳腺癌MCF-7细胞迁移的影响。结果 MTT实验结果发现,不同浓度的蜜炙紫菀水煎剂(25、30、35、40、80 mg/ml)对乳腺癌MCF-7细胞的增殖具有明显的抑制作用,抑制率分别为2.19%、12.58%、14.36%、15.96%、54.69%。流式细胞术的结果显示,与空白对照组相比较,蜜炙紫菀水煎剂可以使乳腺癌MCF-7细胞周期的各个时相(G1、S和G2期)的细胞所占百分数发生改变。浓度为25 mg/ml和35 mg/ml的蜜炙紫菀水煎剂能使G1期中细胞所占的百分数增加,且两个浓度的细胞凋亡率都大于空白对照组。划痕实验结果显示浓度为25 mg/ml和35 mg/ml蜜炙紫菀水煎剂组较空白对照组划痕愈合率偏低。结论不同浓度的蜜炙紫菀水煎剂在体外实验中对乳腺癌MCF-7细胞的增殖具有一定的抑制作用,同时能够将细胞的增殖周期阻滞在G1期,促进MCF-7细胞的凋亡。一定浓度的蜜炙紫菀水煎剂对MCF-7细胞的迁移具有抑制作用,为临床应用中药紫菀治疗乳腺癌疾病提供实验基础。Objective To explore the impacts on the proliferation,cycle,migration and apoptosis of MCF-7 cell in breast cancer in the in vitro experiment with the decoction of honey processed radix asteris(herbal decoction in short)at different concentrations.Methods Using MTT experiment,the impacts of the medicinal treatment were observed on the growth and proliferation of MCF-7 cell in breast cancer.The flow cytometry was adopted to study the impacts of the herbal decoction on the proliferation,cycle and apoptosis of MCF-7 cell.The scratching test was used to observed the impacts of the herbal decoction on the migration of MCF-7 cell.Results MTT experiment results showed that the herbal decoction of different concentrations(25 mg/ml,30 mg/ml,35 mg/ml,40 mg/ml and 80 mg/ml)apparently inhibited the proliferation of MCF-7 cell of breast cancer and the inhibitory rates were 2.19%,12.58%,14.36%,15.96% and 54.69% separately.The results of flow cytometry showed that compared with the blank control group,the herbal decoction changed the cell proportions in each time phase(G1,3 and G2)of MCF-7 cell cycle.The herbal decoction at 25 mg/ml and 35 mg/ml increased the cell proportion at G1 and the apoptotic rate at these two decoctions was higher than that in the blank control group.The results of scratch test showed that at 25 mg/ml and 35 mg/ml,the healing rates in the decoction group were lower than those in the blank control group.Conclusion The decoction of honey processed radix asteris at different concentrations inhibits the proliferation of MCF-7 cell of breast cancer in the experiment in vitro.The herbal decoction at certain concentrations inhibits the cell migration.All of those provide the experimental basis in the clinical application of radix asteris for breast cancer.
