摘  要:目的:评价使用可注射骨髓间充质干细胞(marrow stromal cells,MSCs)膜片修复兔颅骨极限缺损的成骨能力。方法:将体外分离培养扩增的兔MSCs,使用成骨诱导培养液连续培养2周,形成细胞膜片。采用16只大白兔构建兔颅骨极限缺损模型,随机分为两组,每组8只。实验组,颅骨缺损处注射MSCs膜片;对照组,颅骨缺损处不植入任何材料,分别于术后8周和l2周取材,通过大体观察、X线摄片及组织学检查分析其修复颅骨缺损的效果。结果:体外构建的MSCs膜片为细胞-细胞外基质聚合体,茜素红染色见膜片中细胞外基质内有钙化结节沉积,扫描电镜可见膜片内丰富的胶原纤维和矿化结节聚集。细胞膜片植入兔颅骨缺损,8周时形成组织工程骨,12周时缺损得到完全的骨修复,而对照组仅见纤维组织形成。结论:成骨性MSCs膜片具有良好的骨修复能力。Objective The ability of cell sheet from MSCs to heal critical-size rabbit calvarial defect was evaluated. Methods We harvested MSCs sheets using a continuous culture method. Histological examination and scanning electron microscopy were performed to evaluate the osteogenic sheets. The cell sheet was then implanted into a 15mm diameter rabbit calvarial defect. The new bone formation inside the defect was investigated by radiographic, histological, and histomorphometric analyses in 8 and 12 week after operation respectively. Results The cell sheets showed evident mineralized nodules, indicating their in vitro osteogenic potential. Radiographic, histological and histomorphometric analyses revealed that the defect treated with cell sheet showed significant bone formation. Conclusion The cell sheet can enhance bone regeneration in healing critical-size rabbit calvarial defect.
