摘  要:目的 通过使用C3a受体特异性拮抗剂(C3aRA)研究补体C3a-C3aR在三氯乙烯(TCE)致敏小鼠肾脏免疫性损伤中的作用,探讨职业性三氯乙烯药疹样皮炎(ODMLT)肾脏损伤机制.方法 将42只无特定病原体(SPF)级雌性BALB/c小鼠(6~8周)随机分为空白对照组(5只)、溶剂对照组(5只)、TCE处理组(16只)和TCE+C3aRA处理组(16只),其中TCE处理组与TCE+C3aRA处理组根据皮肤致敏试验评分进一步区分为致敏组和非致敏组.取小鼠血清样品采用生化试剂盒检测小鼠肾功能变化;采用实时荧光定量多聚核苷酸链式反应(qPCR)法检测小鼠肾脏组织中C3aR基因表达水平变化;采用免疫组化法检测小鼠肾脏组织中白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)蛋白表达水平变化.结果 与溶剂对照组和相应非致敏组比较,TCE致敏组和TCE+C3aRA致敏组小鼠肌酐(CRE)和尿素氮(BUN)水平明显升高,差异均有统计学意义(P<0.05);与TCE致敏组比较,TCE+C3aRA致敏组CRE和BUN水平明显降低,差异均有统计学意义(P<0.05).与溶剂对照组和TCE非致敏组比较,TCE致敏组小鼠肾脏组织中C3aR基因表达水平均明显升高,差异均有统计学意义(P<0.05).TCE致敏组和TCE+C3aRA致敏组小鼠肾脏组织中IL-1β和TNF-α蛋白有大量表达,与TCE致敏组比较,TCE+C3aRA致敏组肾脏组织中IL-1β和TNF-α蛋白表达水平明显降低,差异均有统计学意义(P<0.05).结论 补体C3a-C3aR可能参与TCE致敏小鼠肾脏免疫性损伤,C3aRA对TCE致敏小鼠肾脏免疫性损伤有一定的保护作用.Objective To explore the effect of complement C3a-C3a receptor in the kidney immune injury in trichloroethylene-sensitized mice by using C3a receptor specific antagonist C3aRA and discuss the pathogenesis of kidney injury in occupational dermatitis medicamentosa-like of trichloroethylene (ODMLT).Methods 42 female 6~8 weeks old BALB/c mice of specific pathogen free were randomly divided into blank control group (5),solvent control group (5),TCE treatment group (16) and TCE+C3aRA treatment group (16).The TCE treatment group and TCE +C3aRA treatment group were further divided into the sensitized group and the non-sensitized group according to the skin sensitization test score.Renal function was detected by biochemical detection kit;expression of C3aR in kidney tissue was detected by qPCR;expression of IL-1β and TNF-α protein were detected by immunohistochemical.Results Compared with solvent control group and corresponding non-sensitized group,CRE and BUN in TCE sensitized group and TCE+C3aRA sensitized group were significantly increased (P〈0.05).Compared with TCE sensitized group,CRE and BUN in TCE +C3aRA sensitized group were significantly decreased (P〈0.05).Compared with solvent control group and TCE non-sensitized group,the expression level of C3aR gene in kidney tissue in TCE sensitized group was significantly increased (P〈0.05).There was a large number of IL-1β and TNF-α protein expression in kidney tissue in TCE sensitized group and TCE +C3aRA sensitized group.Compared with the TCE sensitized group,thc cxpression level of IL-1β and TNF-α protein in kidney tissue in TCE+C3aRA sensitized group was significantly decreased(P〈0.05).Conclusion C3a-C3aR maybe involved in the kidney immune injury in TCE sensitized mice,C3aRA has a protective effect on the kidney immune injury in TCE sensitized mice.
