摘 要:目的采用LDH法测定纤维蛋白胶对人脂肪干细胞的细胞毒性。方法取成人吸脂术后的废弃脂肪组织并从中得到人脂肪干细胞,分别制成50%浸提液组、100%浸提液组、阴性对照组、阳性对照组,并采用LDH检测试剂盒测定各组纤维蛋白培养液中LDH的含量。结果原代ASCs生长缓慢,传代后生长加速,且ASCs呈成纤维样细胞形态;24 h和72 h两个时间点中50%纤维蛋白胶浸提液组、100%纤维蛋白胶浸提液组分别与阴性对照组比较,差异无统计学意义(P>0.05)。结论纤维蛋白胶对ASCs无明显细胞毒性,可为以纤维蛋白胶为支架材料的脂肪组织工程研究提供参考。Objective To determine the cytotoxicity of fibrin glue to human adipose stem cells by LDH method. Methods The abandoned adipose tissue of human liposuction was taken and the human adipose stem cells were obtained and made into the 50% leaching solution group, 100% leaching solution group, negative control group and positive control group.And the LDH test kit was used to detect the content of LDH in each fibrin culture medium. Results The growth of primary ASCs was slow. The growth of ASCs was accelerated after passage, and ASCs appeared fibroblast-like morphology. There was no significant difference between 50% fibrin glue extract group ,100% fibrin glue extract group and the negative control group at 24 and 72 hours(P〉0.05). Conclusion Fibrin glue has no obvious cytotoxicity on ASCs, and it can give a reference to the study of adipose tissue engineering with fibrin glue as scaffold.
