摘 要:目的探讨极低频电磁场长期暴露对大鼠肺组织的损伤效应。方法 Wistar雄性大鼠32只随机分为暴露组和对照组,采用50 Hz、400μT极低频电磁场进行连续暴露60 d,分别于暴露后6 h、15 d、30 d活杀取肺组织,制作切片,HE染色观察肺部病理变化,免疫组织化学SP法检测肺组织中Iba-1的表达并进行定量分析。结果极低频电磁场暴露后6 h,全肺弥漫性出血,15 d和30 d仍可见出血,较6 h减轻,15 d和30 d可见局部肺泡壁增厚,肺泡腔缩小,于上述3个时间点均可见巨噬细胞显著增多。Iba-1的定量分析结果显示,其积分光密度于暴露后6 h、15 d和30 d均显著增高,但上述3个时间点之间无显著差异。结论极低频电磁场长期暴露可致大鼠肺组织损伤且其Iba-1表达增多,表明肺巨噬细胞参与极低频电磁场所致肺损伤。Objective To explore the effects of long-term extremely low-frequency electric magnetic fields(ELF-EMF) exposure on rats' lungs. Methods 32 male Wistar rats were randomly divided into exposure group and control group. Rats in exposure group were exposed to ELF-EMF(50 Hz,400 μT)continuously for 60 days. 6 h,15 and 30 d after the termination of ELF-EMF exposure,5 rats fromeach group were sacrificed and lungs were collected. Slices of lungs were prepared and HE staining was made then the pathological changes of lung were observed by microscope. The expression of Iba-1 in lung was detected by IHC and image analysis. Results 6 h after the termination of ELF-EMF exposure,diffuse hemorrhage could be seen in whole lung,15 d and 30 d after the termination of ELF-EMF,hemorrhage also could be found but was less serious than that of lungs at 6 h,local thickening of alveolar walls and alveolar space narrowing could be found on 15 d and 30 d after the termination of ELF-EMF.Macrophages increased obviously at these three time point. The image analysis of Iba-1 in lung showed that the integral optical density of Iba-1 were significantly increased on 6 h,15 d and 30 d,but there was no statistical difference among these three time points. Conclusion ELF-EMF exposure(50 Hz,400 μT,60 days) could cause pulmonary injury and increasement of Iba-1,which indicated that macrophases play a role in the pathogenesis of ELF-EMF exposure induced lung injury.
